ObjectiveTo study the effect of platelet-derived growth factor (PDGF)-BB in different concentrations on the proliferation of human endometrial epithelial cells in vitro.MethodsStromal and epithelial cells were separated by enzymatic digestion, mesh filtered and selective adherence method. The isolated cells were cultured and identified in vitro.The confluent cells were digested with trpsine and EDTA.Part of the cells was cultured by cover slip method,while the other part cultured in different concentrations of PDGF-BB(1,10 and 20 ng/ml) for 48. The proliferation of cells were detected by MTT test.ResultsThe isolated cells were identified to be epithelial and stromal cells as they were vimentin and cytokeratin staining positive,the positive ratio reach 95%and 97%. Compared with that of control group, the absorbance values of other groups were all increased (P<0.05 or P<0.01). Besides, the absorbance value rose gradually with the increase of the concentration of PDGF-BB on.ConclusionPDGF-BB can promote the proliferation of endometrial epithelial cells.
