ObjectiveStainless steel material is the most widely bio-medical application of metallic materials up to clinice.In recent years, the development of nickel free high nitrogen stainless steel used for the human body have to pass a series of biocompatibility testing. In this paper, observed by AO/EB staining fluorescence microscopy of nickel-free stainless steel caused by the new medical L929 mouse fibroblast apoptosis in living cells, apoptotic cells and necrotic cells in morphology, their biocompatibility testing.Materials and Methods1.Cells cultureCultivated L929 cells with containing 10%fetal calf serum RPMI 1640 cell medium at 37℃,5%CO2 incubator, and passaged.2. Observations of cell morphologySet the experimental group and control group,In which 5 kinds of metal extraction fluid and containing 10%fetal calf serum RPMI 1640 cell culture medium were cultured L929 cells in 48 hours,under AO/EB staining Observed living cells, apoptotic cells, necrotic cells by fluorescence microscope, and calculate apoptosis rate of each group.ResultFluorescence microscope, choose the apoptotic cells exist within the field, quantity of cell apoptosis cells to early apoptotic cells, seen orange, clumped chromatin nuclei broken into the Advanced cell apoptosis occasionally, and visible apoptotic body, then calculate the mean apoptotic rate in each group are 34.20%.48.03%,29.94%. 52.02%.27.58%andl2.60%, The statistical analysis is P<0.01.ConclusionNickel austenitic stainless steel biocompatibility between Austenitic stainless steel (317L) and co-cr alloy.Experimental metal biocompatibility from good to bad is:Au alloy>Co-Cr alloy> Nickel-free stainless steel>Austenitic stainless steel(317L)>Ni-Cr alloy.
