Objective:Through establishing the HPLC-DAD chromatographic fingerprint analysis method of Dachengqi decoctions and rat serum after administrating its decoctions, to investigate the serum pharmaco-chemistry of Dachengqi decoctions. So it can illuminate the blood transitional components source of Dachengqi decoctions initially, and to a certain extent, it can explain the compound compatibility discipline and effective material basis of Dachengqi decoctions.Methods:First of all, a HPLC-DAD chromatographic fingerprint analysis method of Dachengqi decoction and its decomposed formulas was established, which is used to describe the main information of the components to ensure the quality stability of drug samples. Secondly, the established fingerprints are used to analysis the blood transitional components of rats after administrating the Dachengqi decoction and its decomposed formulas. Then testing the preparation methods of serum sample and establishing the chromatographic fingerprint of blood transitional components of Dachengqi decoctions. According to analysis the fingerprints of vivo and vitro compounds, definite the source of transitional components in rats blood. Then comparing the fingerprints of Dachengqi decoction, Dachengqi decoction lacking of Glauber's salt, compatibility of two traditional Chinese medicines and each single herb, which is to determine the source of transitional components in blood and clarify the principal of compatibility of herbs. Finally, according to the characteristics of the serum transitional components, research some special serum components selectively, so as to finding their metabolic features at different times after administrate the formulas.Results:There are 58 ingredients can be marked in the fingerprint of Dachengqi decoction in vitro, while there are 37 transitional components in rat's serum,29 components of which are from the prototype compound ingredients, the other eight are metabolites and new produced material. It has 25 components in the serum of rat which administrated the single herb Rhubarb, while 10 in Magnolia cortex,15 in Immature Orange Fruit. The main transitional ingredients of Dachengqi decoction are in principal drug Rhubarb, which corroborate that Rhubarb plays a important role in the compound. The combination of Magnolia cortex and Rhubarb in the serum has 26 main ingredients,15 ingredients of which are from Rhubarb,2 from Magnolia cortex,5 can be detected in both single herb Rhubarb and Magnolia cortex. Rhubarb and Magnolia cortex each have 4 ingredients disappeared, and another 4 are new ingredients. The combination of Rhubarb and Immature Orange Fruit in the serum has 27 main ingredients, including 9 ingredients derived from Rhubarb,5 from Immature Orange Fruit,6 can be detected in both single herb Rhubarb and Immature Orange Fruit. There are 8 ingredients disappeared in Rhubarb,2 disappeared in Immature Orange Fruit, and another 5 are new ingredients. The combination of Magnolia cortex and Immature Orange Fruit in the serum has 24 main ingredients, including 2 ingredients derived from Magnolia cortex,7 from Immature Orange Fruit,8 can be detected in both single herb Magnolia cortex and Immature Orange Fruit, and another 7 are new ingredients. When determining some special serum transitional components of rat blood which is collected from the ophthalmic vein, the distribution of the definitive component in rats, that have different administrating, has a regular variation characteristic. The chromatographic peak area increased firstly and then decreased along with the lengthening of the administrate times.Conclusion:This method of serum pharmaco-chemistry is applied to research the material basis of Dachengqi decoction. The results can determine the source of the serum transitional compositions initially. To a certain extent it can explain the compatibility discipline of Dachengqi decoction, and laid a foundation for further studying the material basis of Dachengqi decoction.
