Objects:1. To observe the effects of electro-acupuncture (EA) in different interval times on the sleep time of Pentobarbital sodium rats.2. To observe the effects of electro-acupuncture in different Interval times on the content of Monoamine Neurotransmitter such as 5-Hydroxytryptamine (5-HT),5-Hydroxyindole-3-acetic acid (5-HIAA), Dopamine (DA), Norepinephrine (NE) and Cytokines such as IL-1βin the hypothalamus of insomnia rats caused by DL-P-Chlorophenylalanine (PCPA) to investigate the best interval times and give certain evidences for seeking reasonable treatment interval time in clinical medicine.Methods:1.48 SD rats were divided randomly into control group and EA group. Selected 6h,12h and 24h as treatment interval times. The sleep time of Pentobarbital sodium rats was recorded to observe the changes of sleep time of rats in each group.2. Totally 72 SD rats were divided randomly into control group, model group, EA group. The model of insomnia rats were established by PCPA intraperitoneal injection. Start the treatment after the model was made at the third day. The acupoints of Shenmen (HT7) and Sanyinjiao (SP6) were also chosen, and selected 6h,12h and 24h as treatment interval times,6 day duration. Then 5-HT,5-HIAA, DA, NE and IL-1βcontent detection were observed by Fluorescence spectrophotometry (FS) and Enzyme-Linked Immunosorbent Assay (ELISA).Results:1. Compared with control group, the sleep time of Pentobarbital sodium rats was obviously prolonged in EA groups(P<0.05 or P<0.01);contrasted with each interval times in EA groups, the groups of 6h,12h were better than 24h(P<0.05,P<0.01),obviously prolonged the sleep time of Pentobarbital sodium rats. There was no difference between 6h and 12h.2. After PCPA intraperitoneal injection the content of 5-HT and 5-HIAA in the hypothalamus of rats obviously decreased with comparison to control group (P<0.01), but the content of DA, NE and IL-1βdid not change much compared with control group (P>0.05).3. In the EA groups the content of 5-HT,5-HIAA and IL-1βin the hypothalamus of rats increased obviously in contrast with the model groups (P<0.01), while the content of DA and NE decreased obviously in contrast with the model groups (P<0.01);contrasted with each interval times in EA groups, the groups of 6h,12h were more obvious than 24h on the index above mentioned (P<0.05,P<0.01),There was no difference between 6h and 12h groups.Conclusion:1. Treatment with EA in different interval times is attended with good effect.2. The effects of EA with 6h and 12h interval times increase 5-HT,5-HIAA and IL-1β, decrease DA and NE in the hypothalamus of rats have a better result than 24h.The results indicated that repetition treatment by every 6h or 12h can increase the therapeutical effect obviously. In accordance with the clinical conditions, repetition treatment by 12h is reasonable.
