AIM:To study effects of peripheral blood and expression of blc-2 and caspase-9 mRNA treated by Liuwei Dihuang decoction, Buzhong Yiqi decoction and Fufang Danshen decoction in immunosuppressive mice induced by cyclophosphamide (CTX).METHODS:100 NIH mice were randomly divided into 5 groups, Group A, treated with Liuwei Dihuang Decoction; Group B, treated with Buzhong Yiqi Decoction; Group C, treated with Fufang Danshen Decoction; Group D, the control group, treated with normal saline; and Group E is the Normal Group. In addition to group E, all groups were given CTX by intraperitoneal injection to create the immunosuppressive model. The mice were killed after 14 days and 28 days, and then detected the expression of bcl-2 mRNA and caspase-9 mRNA by RT-PCR.RESULTS:1.After 14 days, the content of peripheral WBC, RBC, HGB and PLT in Control Group were lower than Normal Group; and as compared with Normal Group, the expression of blc-2 mRNA were lower and caspase-9 mRNA were higher in Control Group. They both have statistical significance (P<0.05).2. After 28 days, the content of peripheral WBC, RBC, HGB and PLT in Control Group were lower than Normal Group, and the expression of blc-2 mRNA were lower and caspase-9 mRNA were higher than Normal Group; and as compared with Control Group, the expression of blc-2 mRNA were lower and caspase-9 mRNA were higher in three Treatment Groups; the expression of caspase-9 mRNA in Liuwei Dihuang Group were higher than the other Treatment Groups. They all have statistical significance (P<0.05).CONCLUSION:1.The three decoctions all could increase the content of peripheral blood WBC, RBC, HGB and PLT in immunosuppressive mice induced by cyclophosphamide. 2. Liuwei Dihuang decoctions, Buzhong Yiqi Decoction and Fufang Danshen Decoction as the representative of the Nourish kidney, tonifying spleen and invigorate the circulation of blood all could elevate the mRNA expression of bcl-2 and inhibit the mRNA expression of caspase-9 in splenocytes in immunosuppressive mice induced by cyclophosphamide. It's may be one of mechanism which Chinese traditional medicine inhibit the normal cells from apoptosis in chemotherapy patients.3. Compare with 1 week, Give these three decoctions 4 weeks couldn't increase the content of peripheral WBC, RBC, HGB, PLT, and could not elevate the mRNA expression of bcl-2 and inhibit the mRNA expression of caspase-9, may be the treated time is too short, and the superiority of long-term drug-use of Chinese Traditional Medicine can not be reflected in this experiment, we will prolong the experimental period in later research.4. In Liuwei Dihuang decoctions, Buzhong Yiqi Decoction and Fufang Danshen Decoction as the representative of the nourish kidney, nourish spleen and invigorate the circulation of blood can become a possible tumor patient's adjuvant therapy after chemotherapy.
