The Experimental Study On Hemoperfusion And Hemofiltration For Dogs With Acute Hepatic Failure

Objective:Set up dog's acute liver failure model, study the mechanism of Non-biological artificial liver technology hemoperfusion combined with hemofiltration therapy cure drug-induced acute liver failure, seek new breakthroughs about combined treatment.Methods:Select five female beagle dogs and five male Beagle dogs,the ages of the dogs are 10-12months (13.3±1.18 kg).Randomly divided them into experimental group and control group. Firstly,inject 30% of the acetyl amino phenol (0.5ml/kg)by vein,then inject 40% of carbon tetrachloride(0.5ml/kg) by abdominal subcutaneous.12 hours later,repeat the above medication, but the medication dose is 1/2 of the first administration. The hemoperfusion combined with hemofiltration treatment were given to the treatment group after 24 hours, the control group were not given any treatment. Dynamic monitoring the two groups'general conditions, blood(RBC,WBC,PLT),biochemical indicators (ALT,AST,TBIL,CHE,Cr,BU,TP,ALB,NH3,electrolyte),PT,SOD,MDA. Compare the survival time the survival rate at different time points.Results:The treatment can improve the general state of animals.Compared with the control group,the treatment group's biochemical indicators,PT,SOD and MDA have statistics meanings at the same time points(p<0.05-P<0.01). The treatment group's survival time are longer than the control group,Statistical data:the treatment group (96.1士11.75)h; the control group (47.55士1.89)h,(p<0.05). Survival analysis show the treatment group's survival curve is higher than the control group,Statistical data show that the survival rate have statistical significance (P<0.05).Conclusions:Non-biological artificial liver technology hemoperfusion combined with hemofiltration therapy can prolong acute hepatic failure models'Survival time,improve biochemical indicators,coagulation, increase the survival rate,ease the liver's Lipid peroxidation,and ease the liver's damage.Its therapeutic effect is positive.This combination therapy is a new approachto cure acute liver failure.
