Objective:To establish the female model of kidney essence insufficiency, and the pregnant female model of kidney essence insufficiency by the method of cat horror injuring kidney. The aim is to explore ZSF on the mechanism of Filling Kidney essence.Method:1.72 healthy female Kunming mice were divided into 6 groups: normal group, model group, JSW group, high dosage group of Additive ZSF, middle dosage group of additive ZSF and low dosage group of Additive ZSF(n=12 in each group). Except normal group, other 5 groups were created the model of kidney essence Insufficiency by the method of horror injuring kidney for 24h daily for 21 days. Model group was administered daily distilled water, others were administered daily JSW to JSW group, to ZSF high, Moderate and low doses groups respectively after horror 21 days. Then observed the general situation of animals, measured weight of the body, organ weight, organ index and were detected with radio-immunity the quantities of Estradiol (E2), Follicle Stimulating Hormone (FSH) and Luteinizing hormone (LH) in serum.2:27 males and 54 females healthy Kunming mice, mice were 2:1 matched-pairs with health female mice for 4 days before model making, The pregnant mice were randomly divided into normal group, model group and ZSF group (n=12 in each group). During gestational period, model group were given distilled water, treatment group was given Additive ZSF for 19-21 days. Except normal group, other 2 groups were designed to make model of kidney essence Insufficiency by the method of horror injuring pregnant mice during gestational period for 19-21 days. The mice in model group and ZSF group were frightened by the cat for 24h daily for 19-21 days. Pregnancy rate and average baby counts of every female mice (sum of babies/number of mother mice).The results were examined at postnatal 1 day and postnatal 40,60,75 day. The number, the data of growth and development of the neonatal mice were observed:the weight of body and turn ability neonatal mice for neonatal mice Id, Climbing pole test for neonatal mice 40d,60d,75d, Climbing grid test 40d,60d,75d, Roadway looking for food test 40d,60d,75d, Roadway looking for food test insert cat's quack 40d,60d,75d, Mother mice and 75d neonatal mice were taken the eyeball for blood and separate the serum, were detected with radio-immunity the serum Interleukin-1, Interleukin-2 for mother mice and 75d neonatal mice, measured with GuoFeng method RBC-C3bRR, RBC-ICR for mother mice and neonatal mice 75d and the weight of immune organs.Result:1:the prescription could also effectively increase the content of E2, FSH, LH. Every dosage group of Additive ZSF took effects, especially the high group.2: ZSF prescription may improve pregnancy mice and average baby counts of every female mice, and enhance growth and development of neonatal mice, and may improve Mother mice and neonatal mice the function of red cell immune adherence.Conclusion:Experiments using "horror injuring kidney" and "horror hurt pregnant mice" approach successfully replicated the essence deficiency model. ZSF prescription might ameliorative effect on the reproduction endocrine and immune system function. ZSF prescription may improve pregnancy mice and average baby counts of every female mice, and enhance growth and development of neonatal mice, and may improve Mother mice and neonatal mice the function of red cell immune adherence.
