Objective:Microsporum canis is a zoophilic dermatophytes that most commonly infect cats,dogs and human.It is the primary agent of tinea capitis in humans,especially in children.Recently,the frequency of M.canis infection increased gradually due to the notable growth in the numbers of house pet.This study will focus on one of the several proteases produced by M.canis,which are considered to play a role in the pathogenesis of tinea capitis.By using semi-quantitative RT-PCR,expression level of FSH1 will be detected in the 30 strains of tinea capitis and 30 strains of tinea corporis, and their generations in the Sabouraud liquid medium,and whether they inducted by child's scalp tissue and glabrous skin tissue or not. On the basis of these results,we will study the function of FSH1,which may be helpful for us to find the pathogenic mechanism of tinea capitis caused by M.canis.Methods:30 tinea capitis strains and 30 tinea corporis strains were incubated on rice medium for 2 weeks,and then Five million spores were separated and cultivated in 100 ml Sabouraud liquid medium for 21-28 days, and mycelium masses were collected as the first generation,the subsequent five generations were collected in the same way.Meanwhile,the same number of spores were cultivated in 100 ml mineral medium with child's scalp tissue and glabrous skin respectively,and the mycelium masses were collected 21-28 days later.Semi-quantitative RT-PCR was used to analyze the expression of FSH1 among the above strains.Results:M.canis grew well and produced mycelium masses in the Sabouraud liquid medium or mineral medium supplemented with children's scalp tissue or glabrous skin.Comparatively,they grow better in Sabouraud liquid medium than in the others.We applied semi-quantitative RT-PCR to detect the expression of FSH1 in M.canis,and the results showed that the FSH1 was expressed in all strains. The expression between the strains of tinea capitis and tinea corporis different significantly (P<0.01),but there is no statistic difference between the strains of their generations in the Sabouraud liquid medium (P>0.05).There was significant difference in the FSH1 level before and after the indution by tegument tissue (P<0.01),between the strains inducted by the child's scalp tissue and glabrous skin tissue is obvious (P<0.01).Conclusion:1.FSH1 was actively expressed in M.canis.2.A higher expression levels were found in the strains of tinea capitis compared with the strains of tinea corporis.3.Their generations in Sabouraud liquid medium expressed FSH1 constantly.4.The FSH1 expressed in the strains induced by tegument tissue is much more obviously than the strains not induced,and the strains inducted by child's scalp tissue is the most obviously.5.FSH1 is probably related to the pathopoiesis of tinea capitis caused by M. canis.
