Objective: To observe the carrying targeted angiopoietin-2 gene in the lentiviral siRNA on cultured human pancreatic cancer PANC -1 cells, angiogenesis and invasion effects.Methods :LV-Ang2-siRNA transfected into human pancreatic cancer cell line PANC-1 in green fluorescent protein (GFP) gene as reporter gene, using fluorescence microscopy and flow cytometry virus transfection efficiency. Reverse transcription-polymerase Chain Reaction Detection of PANC-1 cells, Ang2-mRNA and protein expression, MTT method and flow cytometry cell cycle and apoptosis, embryo, and invasion assay detected the transfer of tumor angiogenesis and invasion ability.Results :LV-Ang2-siRNA was successfully transfected into PANC-1 cells, FCM determination of transfection efficiency of around 95%. With the negative control group and blank control group, experimental group PANC-1 cells, LV-Ang2 mRNA and protein expression was significantly inhibited. The experimental group significantly decreased cell transfer and the invasiveness of tumor angiogenesis significantly reduced, the differences were statistically significant ( P <0.05). The apoptosis rates of the three groups were lower, no significant difference between the groups statistically significant (P> 0.05). Conclusions :LV on PANC-1 cells transfected with a better ability to be mediated gene expression in PANC-1. LV-Ang2-siRNA transfected PANC-1 cells can reduce the PANC-1 cell line Ang2mRNA and protein expression significantly reduced the invasive ability of tumors and inhibit tumor angiogenesis, However, the inhibition of their proliferation and apoptosis had no significant effect.
