| Objective: To observe the effects of intra-articuar injection with COX-2 inhibitor, HA and the compounds of them to articular cartilage and synovial membrane on osteoarthritis. Methods: Thirty-five New Zealand white rabbits were randomly divided into 5 groups: the negative control group (group A),the compound of COX-2 inhibitor and HA group (group B),the HA group (group C),the COX-2 inhibitor group (group D) and the positive control group (group E), each group contained 7 rabbits. The rabbits of group A as the negative control, did not received any treatment; from group B to group E, after construct OA model with Hulth method, we took administration of intra-articular injection with 0.3ml of the compounds,HA,the COX-2 inhibitor and saline once a week for five weeks. MRI examination were taken 13 weeks later, and the blood and synovial fluid samples were taken from each group for IL-1 detection, the right knee specimens were taken from each group for general observation,Mankin score,histopathological observation. The statistical analysis of the data was performed with SPSS 17.0.Result:①M ankin score: increased gradually from group A to group E, there was significant difference between the groups (χ2=29.82,P<0.01)and pairwise comparison between any two groups (P<0.01).②Optical microscope: pathological results showed that cartilage cells gradually decreased in disorder from group A to group E, articular cartilage surface was progressively fibrosis and thinning, the tide-line became disorder and some of subchondral bone was exposed.③The level of IL-1: the level of the group B or C or D or E was higher than the group A. From group A to group E, it increased gradually. Between the groups there was statistically significant difference (Fsynovial fluid =2217.20,Fserum =1594.48,P<0.01), the trends were same in the synovial fluid and serum , the level of IL-1 in synovial fluid was higher than that in the serum.④MRI: From group A to group E, the changes of the outline of cartilage and cartilage signal became more clear, followed by the increasing in joint effusion, synovial hyperplasia, there was a series of secondary changes in joint space and subchondral bone.Conclusions:①OA animal model, which made by Hulth method, can be used to observe the patholgical changes in the research of the intra-arthicular drugs therapy on OA.②Using liposomes as a carrier, we can manufacture a stable compound of COX-2 inhibitor-HA which can be applied in the research of degeneration of cartilage and synovial membrane.③IL-1 ,which could reflect the changes of OA, was related with the immunity and inflammatory of OA patients.④MRI examination can be used to study the degeneration of cartilage and synovial membrane in OA model of rabbits.⑤It was more effective than COX-2 inhibitors or HA in preventing or mitigating the pathological changes of OA by appling the compound of COX-2 inhibitor and HA. |
