Objective:The use of animal experiments, through the X-ray irradiation SD rat eyes set up animal model of radiation cataract were observed by X-ray radiation dose and the severity of cataract relations; To detect the expression of AQP1 in the lens and to discuss the relationship between radiation and cataract formation.Methods:Select 8 pre-irradiation of rats as normal control group, experimental exposure to rats 24, the average is divided into 3 groups each group 8, one-time irradiation, respectively (both eyes simultaneously irradiation) 5 Gy,15 Gy,25 Gy. Experimental control group of 24 were divided into 3 groups each of 8, 5 Gy,15 Gy,25 Gy, respectively, as a control group of sham irradiation (irradiation of light in the non-radiation, the other conditions were the same as corresponding to the irradiated group). Rats were randomly check each eye for the study as the experimental eye. All rats were irradiated 1,3,5,7,15,30,45,60,90d, intraperitoneal injection of 3.6% chloral hydrate anesthesia, the use of prescribed eye drops to fully mydriasis, slit-lamp microscope and record the lens opacities case; in 3 months after irradiation the rats were sacrificed, check the lens for homogenate, select the methods of immunohistochemistyr and RT-PCR to detecte the AQP1 expression.Results:Histology:The observation of 3 months, normal control group, the experimental control group and 5Gy rats no abnormal lens. 15Gy group 45d after irradiation and after posterior capsular cortex began to appear small, scattered spots in the white turbidity, and then blended into more labor-intensive, non-uniform punctate, patchy opacities, the further development of a more even after the emergence of dense cortical opacities, the former cortex , anterior capsule opacification and mild core. One of 3 months up to 62.5% in stage â…¡, 37.5% for stageâ…¢. 25Gy group similar changes, but the turbidity of time crystal happened sooner, faster, more obvious turbidity. 30d after irradiation that is, posterior capsular opacification spots began to appear, since the rapid development of lesions, to 45d when there is 37.5% of all crystal turbidity. To 3 months time in addition to Example 1 under the clear posterior capsule opacification, the remaining seven cases are fully turbid lens.Immunohistochemiistry and computer image analysis show that:AQP1 express in the anterior lens capsule of the lens epithelial.A varying degree of brown-yellow granules can be seen in the membrane.Each group can be seen their expression.With the increased exposure,AQP1 of all experimental groups are decreased compared to the control group.The difference is significant.( P<0.05)AQP1 expression in all experimental groups are decreased with the dose increase, The difference is significant.( P<0.05) RT-PCR and computer image analysis show that:At the mRNA, AQP1 express in the anterior lens capsule of the lens epithelial. Positive AQP1 gene can be carried out byRT-PCR. With the increased exposure,AQP1 of all experimental groups are decreased compared to the control group.The difference is significant.( P<0.05)As the exposure increase, the AQP1 expression are reduced in dose-dependence, The difference is significant.( P<0.05)Conclusions:High-dose X-ray in the short term can cause lens opacities; The speed of radioactive cataract formation has a positive correlation with the radiation dose.In the radiation-induced cataract model,the expression of AQP1 in lens change.Radiation may reduce the expression of AQP1 in lens epiyhelial cells,affect the water metabolism of lens.AQP1 may play an important role in the formation of cataract.
