Objective :To study the inhibitory effects and its mechanisms of Ori on Human pancreatic carcinoma SW1990 Cells .Methods :The expression of survivin,Bax and Bcl-2 protein in pancreatic carcinoma was examined by immunocytochemistry . The inhibition of cell growth mediated by Ori on SW1990cells was measured by MTT assay . The morphological changes were observed by Hoechst33258 fluorochrome staining and electron microscope. Cell cycle and apoptosis rate were analysed by flow cytometry. The mRNA expression of survivin,p53,p21,cyclinB1,cdc2,bcl-2,bax and cytochrome c after the Ori treated on SW1990cells were studied by RT-PCR .Results :The expression of Survivin, Bax and Bcl-2 protein in pancreatic carcinoma were upregulated and closely correlated with the clinical stage and metastasis of pancreatic carcinoma. The growth of humen pancreatic carcinoma SW1990 Cells was significantly inhibited by Ori . Apoptosis morphological changes about chromatic agglutination and nuclear condensation were detected by Hoechst 33258 fluorochrome staining and electron microscope in Ori treated SW1990 cells."Sub-G1"phase peak and G2/M growth arrest were found using flow cytometry. Apoptosis rate of SW1990 cells treated with 10,20,40μmo/L Ori for 24 h were 1.84%,5.41%(P<0.01),16.56%(P<0.01), respectively.The upregulating mRNA expression of p53,p21,bax,cytochrome c and downregulating mRNA expression of survivin,cyclinB1,cdc2,bcl-2 were detected by RT-PCR.Conclusion The inhibitory effect of Ori on Human pancreatic carcinoma SW1990 Cells through induced apoptosis and G2/M growth arrest and the mechanisms may be associated with uprugulating mRNA expression of p53,p21,bax,cytochrome c and downregulating mRNA expression of survivin,cyclinB1,cdc2,bcl-2.
