Objective:Investigating the protective effects of tea polyphenols on testis in injury induced by establishment of experimental animal model of testicular torsion/detorsion.and further to investigate The mechanism of damage to torsional testicular and spread medication of testicular torsion.Methods:Thirty two healthy male Wistar rats were randomly divided into 4 Group,each comprising 8 ra ts.In GroupⅠ,rats' left scrotal incision,free exposed testes,arbitrarily moved imitated by Turner model,but not reversed,then sutured the scro-tum. In GroupⅡ,rats were tested to left lateral 720°testicular torsion following Turner'method:sutured of tunica albuginea to prevent spontaneous detorsion,then sutured scrotum. then detorsion in six hours.In GroupⅢ,testicular torsion model were established following the same process like GroupⅡ, Intraperitoneal injection of polyphenols(40mg/kg) initiated 30 minutes prior to detortion(5.5h after the torsion) in GroupⅢ. In GroupⅣ,after establishment of testicular torsion model, intraperitoneal injection of polyphenols(40mg/kg) initiated 30 minutes prior to detortion,then low-dose maintenance(10mg/kg per day) in postoperative three days. All the rats were fed under the same condition for 5 days and sacrificed, left torsional lateral testes were harvested for paraffin-embedded sections and homogenate to be examined.To investigate germ cell apoptosis by TdT-mediated d-UTP nick end labeling(TUNEL) spe-rmatogenic apoptosis cell dyed brown. Each sections selected five vision through optical micros-cope(×200), counted 100 cells,and averaged obtained percentage of positive cells apoptosis to statistical analysis, to investigate superoxide dismutase(SOD)and malondialdehyde(MDA) by chemical colorimetric method.All the results are present by x±s, used SPSS 12.0 statistical data software for statistical analysis. Homogeneity test methods used single-factor analysis of variance, size of test wasα=0.05. Result:1 GroupⅢ(testicular torsion/detorsion plus Single Polyphenols of tea injection treatment group) was compared with GroupⅡ(testicular torsion/detorsion group):SOD levels significantly increased and the difference was significant(P<0.05).Content of MDA were reduced drastically and the difference was significant(P<0.05).Apoptosis index was significantly reduced and the difference was significant(P<0.05).2 GroupⅣ(testicular torsion/detorsion group plus Mutiple Polyphenols of tea injection treatment group) was compared with GroupⅡ(testicular torsion/detorsion group):SOD levels were significantly higher and the difference was significant(P<0.05).Content of MDA was significantly reduced drastically and the difference was significant(P<0.05).Apoptosis index was significantly lower and the difference was significant(P<0.05)3 GroupⅣ(testicular torsion/detorsion group plus Mutiple Polyphenols of tea injection treatment group) was compared with GroupⅢ(testicular torsion/detorsion plus single Polyphenols of tea injection treatment group):SOD levels were significantly higher and the difference was significant(P<0.05),but Content of MDA and Apoptosis index were not significantly different(P>0.05)Conclusion:Polyphenols of tea injection could reduce the damage of torsional testicular ti-ssue and apoptosis of the testicular spermatogenic cell; Polyphenols of tea protects torsion/deto-rsion testicular tissue and spermatogenic function through reducing the production of oxygen-derived free radicals and improving antioxidant enzyme activity.
