Objective Discuss the TP15, TP17 TP47 antigen-specific cell-mediated immunity of primary syphilis, secondary syphilis, latent syphilis and two years after the treatment of syphilis patients but whose RPR still negative. Detected if the syphilis patients have existence of consistency in specific cell-mediated immunity and humoral immunity of TP15, TP17 and TP47 antigen.Methods Use TP15, TP17, TP47 three kinds of syphilis antigen-specific as stimulus, stimulate to the healthy, diagnosed primary syphilis, secondary syphilis, latent syphilis and two years after the treatment of syphilis patients but whose RPR still negative in non-experimental group and observation group. Using Elispot technique (enzyme-linked immuno-spot, Elispot) detection of syphilis infection's specific T cell response of peripheral blood volume's measurement to determine specific cell immune caused by TP15, TP17, TP47.Use immuno-slot blot method to detect syphilis patients's specific antibodies TP15, TP17 and TP47 to determine their humoral immunity caused by the situation.Results We use immune-solt bolt to detect syphilis patients's humoral immunity of TP15, TP17 and TP47. The result showed that in primary:positive rate of TP15 is,25%TP17 is 100%,TP47 is 87.5%.The statistically test resultbetween TP15 and TP17 theχ2=4.17, P< 0.05,have statistically significant; between TP17 and TP47 theχ2=0.00, P>0.05,have no statistically significant.In secondary syphilis:positive rate of TP15 is 80%,TP17 and TP47 is 100%. Between TP15 and TP17 or TP47 theχ2=1.75, P>0.05, have no statistically significant.Compare with other group secondary syphilis positive detection rate is the highest. In this study, we use Elispot detect three kinds of Treponema pallidum specific protein. The results showed that compared with the normal group, secondary syphilis have significantly different among the various antigens. Primary, secondary, and serum fixed syphilis patients's TP17 antigen expression was significant difference compared with normal group. Latent syphilis compared with the normal group, have no statistical difference in each three kinds of antigens.Conclusions TP15, TP17, TP47 three kinds of recombinant proteins used as antigen stimulus, Immun-solt blod technology to detect each stage of syphilis patients's humoral immunity and Elispot technology to detect cell-mediated immunity.The result showed that all the three antigens could cause a wide range in specific humoral immunity, but the specific cell-mediated immune response is relatively weak. Our result may provide laboratory evidence for syphilis patients's immune changes in the pathogenesis and some new ideas in research of pathogenesis of syphilis.
