Objective To study the remodelingβcells and its mechanism. rat in modeling period was treat by STZ,and the rats were divided into normal group (control), STZ treated group (STZ), with respect to their blood glucose levels, body weight and pancreas weight of observation statistics. We used double immunofluorescence to observe the subject of some of the key transcription factor differentiation pdx1, ngn3, pax4, Nkx6.1 in the normal group and treatment group, cellular localization, and with western blot to observe differences in the expression of the situation after STZ treatment. The results were observed a transient increase in blood glucose after STZ treatment, insulin structural damage, extreme reduction of isletβcells, and then gradually recovered. Islet ? cells after STZ treatment, the number of substantial increase, and begin to express differentiation marker transcription factor ngn3, quantitative results showed that ngn3 protein expression was significantly increased. Meanwhile, the treatment groups islet ? andβcells were significantly high expression of differentiation transcription factor pax4, while protein expression showed that the absolute increase. Nkx6.1 is only located in the pancreatic isletβcells. Conclusion remodeling in rats after STZ treatment with high expression of ngn3 in the pancreas islet cells ? cells may be pancreatic isletβ-cell source.? ?prompted remodeling of islet?Normal rats cells also expressed pax4, pax4 treatment group was significantly higher expression, this may trigger the expression of ngn3 start with the formation of cells causes the process, it may reshape?isletβcells STZ treated rats with high blood sugar causes hypertriglyceridemia can be automatically restored. Therefore, remodeling of islet ? cell research can be used as treatment for diabetes.
