Objective To find a new cell culture method for cartilage defect repair with the acquisition of a sufficient number of chondrocytes. Methods Human bone marrow mesenchymal stem cells induced differentiation in vitro expansion mixed and cocultured with rabbit mature articular chondrocytes. (RCH) Result Mixed pellet culture showed upregulation of cell proliferation, cartilaginous extracellular matrix production,type II collagen gene and SOX9 expression mesenchymal stem cells differentiate into chondrocytes after 4 weeks. Conclusion MSCs mediated a paracrine effect on BCH through growth factor secretion or cell-to-cell interaction upregulating the cartilaginous phenotype of RCH.
