Part 1 Series diversity in palatal suture of mice during mechanical force-expansionObjective To detect the consecutive diversity in the maxilla palatal suture of mice during the mechanical force-expansion period and discuss the significance of them in the suture remodeling after the expansion.Methods Six week-old male C57BL/6 mice were subjected to palatal suture expansion by opening loops with an initial force of 0.56 N for the periods of 1, 3, 5,7, 14 or 28 days. The sequence of histological changes in the midpalatal suture bone formation of animals was detected by hematoxylin eosin staining and Masson staining.Results Starting at day 1, cells expressing was seen. New cartilage and bone formation was observed at the oral edges of the palatal bones at day 7. the maxilla palatal suture was expansed from the experimental 14d to 28d;compared with the control group, collagen fiber increased significantly along the inner lateral side of the cartilaginous tissue in expansion group from day 1 to day 28 ,and day14 is the maximum.Conclusions These findings demonstrate that expansion forces across the midpalatal suture promote collagen fiber increased significantly which increased proliferation and differentiation.Part 2 CD13/APN and THBS1/TSP-1 in palatal suture of growing mice during mechanical force-expansionObjective To detect the CD13/APN and THBS1/TSP-1 in the maxillary palatal suture of mice during the mechanical force-expansion period and to discuss the significance of them in the suture remodeling after expansion.Methods Six-week-old male C57BL/6 mice were subjected to palatal suture expansion by opening loops with an initial force of 0.56 N for the periods of 1,3,5,7,14 and 28 days. The sequence of CD13/APN and THBS1/TSP-1 changes in the midpalatal suture formation of animals was detected by immunohistochemical S-ABC method and analised by western-blot.Results APN can be found in the mesenchymal cells membrane and the cartilage cells membrane. The expression of APN and THBS1 of expansion group was markedly higher than the control group。APN the highest expression was observed in the 7 day expansion group. THBS1 the highest expression was observed in the 3 day expansion group. The amount of APN and THBS1 expression declined gradually from the 14days expansion group to the 28 days expansion group; however it was still higher than the control group. The expression of APN showed no distinct difference between 1 day and 3days; 14 days and 28 days in the expansion group.Conclusions These findings demonstrated that expansion forces across the midpalatal suture could promote the expression of APN and THBS1, and it might play an important role in the angiogenesis and osteogenesis process.
