Objective To study the effect and mechanism of the proliferation and apop tosis on growth plate chondrocyte of Zinc deficiency in ratMethodsGrowth plate chondrocytes cell were isolated from rib cartilage of 60- to 80-g wista rat. Low zinc cell model was established by TPEN, a kind of chel ating agent which chelates specially to zinc. The relative survival rate were an alyzed by MTT. The rate of the apoptosis cells were analyzed by flow cytome try (Annexin V-FITC/PI). Real-time quantitative PCR and the Western-Blot me thod was used to measure expression levels of ZNT-1 the gene and the protein um expression in zinc depleted Growth Plate Chondrocytes cell on various con centration of TPEN exposure(0,5,10 and 20μmol/L 12h)ResultThe apoptosis cells rate of the Growth plate chondrocytes cells were signif icantly increased when exposed to TPEN. The results show that the apoptosis cells rate reached 9.93% in 0 group,94.78% in 7μmol/L group. Zinc depletion induced by the zinc chelator TPEN (N,N,N',N'-Tetrakis-(2-pyridylmethyl) ethyl ene diamine) resulted in a up to induction of transporters ZnT-1 mRNA levels .The result of the Western Blot show the same trend. ConclusionIt may be that extracellular zinc ZnTl through the regulation of cell prol iferation and thus reduce the growth plate chondrocytes to promote apoptosis.
