Objective:By measuring the content of PDCD4 tumor tissues of esophageal cancer patients we explored the relation between expression of PDCD4 in esophageal cancer and nature of esophageal (level, degree of differentiation, pathological stage, gender, age). Then we analyzed the causes and explored esophageal cancer PDCD4 Clinicopathological significance of PDCD4 expression. We hope this finding will provide clinical treatment and prognosis with new ideas, methods and reference.Methods:Tumor tissue samples from 35 esophageal squamous cell carcinoma patients of Provincial Hospital Affiliated to Shandong University were selected after esophagectomy, while the normal tissues 5cm above the edge of of tumor tissues from 18 cases were regarded as control. All patients did not receive any radiotherapy and chemotherapy treatment before operation. Immunohistochemistry and Western Blot were used to detect PDCD4 expression of 35 cases of human esophageal tumors and 18 cases of normal esophageal tissues.PDCD4 expression levels of PDCD4 expression in esophageal cancer and esophageal cancer in the level and degree of differentiation, pathological stage, gender, and age relationship.Results:1) Immunohistochemical staining showed that PDCD4 expression was 100% in the normal esophageal tissues while it was 42.86% in 35 cases of esophageal carcinoma, the difference was statistically significant (P<0.05). Western Blot analysis revealed that all of PDCD4 protein expression in esophageal cancer tissues were significantly reduced or absent compared with adjacent normal esophageal tissues. (P<0.05) 2) Mean PDCD4 expression of well-differentiated, moderately differentiated and poorly differentiated squamous cell carcinoma were 88.89%,37.5% and 10% respectively. There were significant difference between Well differentiated squamous cell carcinoma and moderately and poorly differentiated squamous cell carcinoma (P<0.05), but there was no significant difference between moderately differentiated squamous cell carcinoma and poorly differentiated carcinoma. (P> 0.05) 3) Mean PDCD4 expression of (â… ,â…¡and III) esophagus carcinoma were 50.0%,43.75% and 38.46% respectively. There were no significant differences among the three groups (P> 0.05).4) PDCD4 expression of esophageal carcinoma from men and women were 48.0% and 30.0% respectively. There was no sexual statistical difference (P> 0.05). PDCD4 expression in esophageal cancer patients of patients over 60 and lower 60 were 33.3% and 52.9%respectively and the difference had no significance (P> 0.05).Conclusion:(1) PDCD4 protein expression in esophageal cancer decreased compared with normal tissue. (2) PDCD4 protein expression in esophageal carcinoma correlated with tumor differentiation. Well differentiated squamous cell carcinoma of PDCD4 protein expression was significantly lower than that in poorly differentiated squamous cell carcinoma and there were no significant difference between poorly differentiated squamous cell carcinoma and moderately group. (3) PDCD4 protein expression in esophageal cancer was not affected by sex, age and TNM staging.
