| Objective:Rehmannia polysaccharide,Plantago polysaccharide,Epimed-ium polysaccharide,dodder seed polysaccharide are all water-soluble components attributed to the liver and kidney meridian,by observing their effects on the growth and differentiation of Adult rat liver stem cell. we can understand the meridian theory of traditional Chinese medicine and verify the theory of the liver and kidney having a common source whether take the Pharmacology as the rationale.Methods:SD rats fed with 3'-methyl-dimethylamino azobenzene (3'-Me-DAB) for 4 weeks,we prepare at liver stem cell from the liver by perfusion digestion,gradient centrifugation method.Through removing non-adherent cell via renewing culture fluid, we can separate and purify the stem cells and then,made them passage and proliferate.Cell morphology was observed under inverted microscope so as to acquire the rule of cell metabolism. Then we observed the effects of different concentrations of Rehmannia polysaccharide,Plantago polysaccharide-e,dodder seed polysaccharide on liver stem cell growth,proliferation and apoptosis,applying tetrazolium salt (MTT Cell Proliferation As-say,MTT assay) to monitor activity of liver stem cells; and introducing RT-PCR to detect HNF4a mRNA expression in adult rat liver stem cells after induction by active ingredients.Results:MTT assay results showed that:specific concentration of Epimedium polysaccharide,dodder seed polysaccharide can increase the activity of liver stem cells, promote cell proliferation. Whereas,certain concentration range of Rehmannia polysaccharide,plantain seed polysaccharide can not promote cell growth and proliferation. RT-PCR results showed that:induced by specific concentration of epimedium polysaccharide and polysaccharide environment,cells HNF4a mRNA expression was significantly increased.Conclusions:(1) Specific concentration of epimedium polysacchar-ide,dodder seed polysaccharide can promote the growth and proliferation of adult rat liver stem cells. A certain concentration range of Rehmannia polysaccharide,Plantago polysaccharide had no effect on adult rat liver stem cells,indicating that polysaccharides cold herb property posted little significant promoting effect on the growth and metabolism of stem cells. (2) Induced by epimedium polysaccharide and polysaccharide,cells HNF4a mRNA expression was significantly increased,suggesting that polysaccharide hot herb property can induce the increase of liver stem cell activity and the tendency to differentiate to liver cells significantly.Keywords:Rehmannia polysaccharide,Plantago polysaccharide,epimedi-um polysaccharide,dodder seed polysaccharide,stem cells,the hepatore-nal homology... |
