| Objective:To study the Clear away Heart and supplement Qi therapeutic method treatment chronic rhinosinusitis and the common molecular mechanisms of disease from the point of view to impact on the inflammatory cytokines by animal experiments.Methods:To take weight 1.5～3.0kg healthy New Zealand white rabbits 100, male and female half and half.Ⅱlevel bio-safety standards by keeping sterile after 1 week, according to the order number into the laboratory, using SAS software, random number table generated randomized, divided into:normal group, model group,sham operation group, traditional Chinese medicine group, western medicine group,5 groups,each group 20. Then reported by Lin Bin, etc. "Do not completely plugged sinus+sinuses lien cotton" in the modeling method,, rabbit model of chronic rhinosinusitis. After modeling 1 day, since the general situation of the animals and to observe changes in symptoms and signs; 43 days after modeling, starting treatment group and western medicine group were fed with corresponding drugs by 1.5ml/kg·d and 25mg/kg·d. After takeing drugs 2 weeks, serum extracted from heart blood, animals were killed, and then set on ice in the ultra-clean out the nasal and sinus mucosa.To detect rabbit serum IL-8, TNF-αlevels by ELISA; the first extract out the nasal and sinus mucosa mRNA, then the mRNA will be transcribed into the corresponding cDNA,to use real time PCR method detecting the expression of tumor necrosis factor-amRNA (TNF-αmRNA). And in the light microscope to observe the pathological changes of the nasal and sinus mucosa.Results:1. Model rabbit serum IL-8, TNF-αlevels and the expression of rabbit nasal and sinus mucosa TNF-amRNA was significantly higher compared with the control group, there were significant differences (P<0.01); 2. Chinese medicine group and western medicine group rabbit serum IL-8, TNF-αlevels and the expression of rabbit nasal and sinus mucosa TNF-αmRNA were lower compared with the model group were significant (P<0.05 or P<0.01), and the normal group no significant difference compared (P> 0.05); 3. sham operation rabbit serum IL-8, TNF-αlevels and the expression of rabbit nasal and sinus mucosa TNF-amRNA compared with the normal group showed no significant difference (P>0.05); 4. light microscope observation showed that:the nasal and sinus epithelial of traditional Chinese medicine group is repaired better, and the hyperplasia of inflammatory cells, glands cell and goblet cell are significantly better than western medicine.Conclusion:The Clear away Heart and supplement Qi therapeutic method can effectively improve the performance of general and local symptoms to caused by chronic rhinosinusitis, and can improve the pathological response in nasal and sinus mucosa. At the same time, can reduce rabbit serum IL-8, TNF-a concentration and the expression of rabbit nasal and sinus mucosa TNF-amRNA to inhibit the development of inflammation and the damage of the nasal and sinus mucosa.
