Objetctiveβ-blocker is a new drug that is treatment of hemangiomas in recent years, but its'mechanism is unclear. The aim of this study was to investigate the expression and significance of PCNA and VEGF onβ-blocker for infantile hemangiomas, and to explore the mechanism of treatment on infantile hemangiomas treated withβ-blocker, so as to provide the theoretical evidence ofβ-blocker in treatment of infantile hemangiomas in clinically.Methods The fresh hemangioma tissue that come from rapid growth, without any treatment and less than 8 months infants were collected. Then put the hemangioma tissue in sterile conditions and general anesthesia, removed the adipose tissue of them, and cut the hemangioma tissue into pieces about 1mm×1mm×2mm. These pieces of hemangioma tissue were cultivated by 1640 nutrient solution including 20% fetal bovine serum with 100μ/ml penicillin and streptomycin in 37℃and 5% CO2 incubator. The groups were divided into the propranolol group, the prednisone control group and the blank control group. In the propranolol group, the propranolol concentration was 10μg/ml. In the prednisone control group, the prednisone concentration was 10μg/ml. In the blank control group, without other drugs in nutrient solution. Hemangioma specimens which cultivated after 8h, 24h, 48h, 72h were collected, then fixed in 4% polyphosphate formaldehyde, embedded, sliced and detected with immunohistochemistry for the expression of PCNA and VEGF of endothelial cells of hemangiomas. The expression image of PCNA and VEGF were analyzed each group by IPP 6.0 software, then statistical analysis was done that used t-test between two groups, and analysis of variance among groups.Results Compared the propranolol group with the prednisone control group and the blank control group after cultivating 8h, the expression of PCNA and VEGF of endothelial cells of Hemangioma in the propranolol group were lower than that in the blank control group(P<0.05), the expression of PCNA and VEGF of endothelial cells in the propranolol group had not different from that in the prednisone control group(P>0.05). After cultivating 24h, the expression of PCNA and VEGF of endothelial cells in the propranolol group were significantly lower than that in the blank control group(P<0.01),the expression of PCNA and VEGF of endothelial cells in the propranolol group had not different from that in the prednisone control group(P>0.05). After cultivating 48h, the expression of PCNA and VEGF of endothelial cells in the propranolol group were significantly lower than that in the blank control group(P<0.01),the expression of PCNA of endothelial cells in propranolol group had not different from that in the prednisone control group(P>0.05),the expression of VEGF of endothelial cells in the propranolol group was lower than that in the prednisone control group(P<0.05). After cultivating 72h, the expression of PCNA and VEGF of endothelial cells in the propranolol group were lower than that in the blank control group(P<0.01), the expression of PCNA of endothelial cells in the propranolol group had not different from that in prednisone control group(P>0.05),the expression of VEGF of endothelial cells in the propranolol group was lower than that in the prednisone control group(P<0.01).Conclusionsβ-blocker(propranolol)can degrade the expression of PCNA and VEGF that are related to the proliferation of hemangiomas. Compared the expression of PCNA and VEGF of endothelial cells on propranolol and prednisone for infantile hemangiomas, propranolol can reduce the expression of VEGF of endothelial cells more obviously than prednisone. It is suggested that propranolol may be inhibited proliferation of hemangiomas through down-regulation of PCNA and VEGF expression, and may be more effective treatment than prednisone.
