| 山西医科大å¦ç¡•士å¦ä½è®ºæ–‡Objective This study was to discuss the effect of Ligustrazine on pulmonary arterial pressure Monocrotaline(MCT) induced pulmonary arterial hypertension and the expression of MMP-2,9, and to discuss the possible affect mechanism of Ligustrazine on pulmonary arterial hypertension and reconstruct of pulmonary arterial structure.Methods Adopting of one abdomen injection in Monocrotaline to bulid pulmonary arterial hypertension.18 Male Wister rats were randomly divided into 3 groups:normal control,pulmonary arterial hypertension model and Ligustrazine treatment groups (n=6 in each group). The fastest flow rate and root of pulmonary artery measured by color Doppler ultrasound. The mPAP and mCAP and the change of right heart plump index was measured, To bserve the pathology alteration of lung tissue by HE staining,the change of MMP-2,9expression in pulmonary was measured. To analyse all date in SPSS 13.0 statistics software.Results (1) Compared with the control group, the fastest flow rate and root of pulmonary artery of the model group and treatment group, increased significantly(P<0.01); treatment group is significantly lower than model group (P<0.01).(2)Compared with the control group, (mPAP)and(mCAP) of the model group and treatment group, increased significantly(P<0.01); treatment group is significantly lower than model group (P<0.01).(3) Compared with the control group, right heart plump index of the model group and treatment group, increased significantly(P<0.01); treatment group is significantly lower than model group (P<0.01).(4) Compared with the control group, pulmonary arteryof treatment and modlegroup increased significantly, the middle of smooth muscle cell proliferaed and the elastic fibers Increased, significantly stenosised. (5) expression of MMP-2,9 in lung tissue: Compared with the control group, MMP-9 expression of treatment and modlegroup increased significantly (P<0.01); Compared with the modle group, MMP-9 expression of treatment group decreased significantly(P<0.01); Compared with the control group, MMP-2 expression of treatment and modlegroup increased significantly (P<0.01).expression of MMP-2 treatment group and model group did not increased significantly (P>0.05).Compared with the control group and treatment group, small artery of model group changed significantly.Conclusion The method of building pulmonary arterial hypertension in MCT abdomen injection singlely is easy and a little damage and high succession; Ligustrazine can reduce pulmonary arterial pressure.; Ligustrazine reduce compound of MMP-9 or decreas activity of MMP-9,this possiblely is one of the affect mechanism. |
