Objective: To study the Effect of GLP-1 analogues on the Expression of PEDF,TGF-β1 in the Kidney of the Rats with Diabetic Nephropath.Methods:1,The rats models of diabetic nephropathy were established by injecting with streptozotocin intraperitoneally. S-D rats were randomly divided into 3 groups: the diabetic nephropath group ,Low-dose treatment group and High-dose treatment group.2,The rats in each group were respectively treated 6 weeks after model Establishment. Blood glucose (BS) ,lipid and 24 h urinary protein (24 h Upr) were determined. Masson staining was used to observe morphoLogical changes of renal tissue.3,Immunohistochemistry and RT - PCR were used to determine the protein and mRNA expression of PEDF,TGF-β1 in renal tissue.4,Statistical analysis: All statistical analysis was performed with Windows SPSS17.0, date are presented X±S. Homogeneity of variance between groups compared with the LSD-t test;. Differences were considered significant at a value of P<0.05.Result:1,Compared with those in the diabetic nephropath group,BS,lipid and 24h Upr deteriorated significantly in the Low-dose treatment group and High-dose treatment group (P<0.05). Compared with those in the Low-dose treatment group,BS,lipid and 24h Upr deteriorated more significantly in the High-dose treatment group (P<0.05).2,Compared with those in the diabetic nephropath group,the protein and gene expression of PEDF increased significantly,TGF-β1 reduced significantly in the Low-dose treatment group and High-dose treatment group (P<0.05). Compared with those in the Low-dose treatment group,the protein and gene expression of PEDF increased significantly,TGF-β1 reduced significantly in the High-dose treatment group (P<0.05).Conclusion:1,The kidney injury in the rats with diabetic nephropathy is ameliorated by GLP-1 analogues.2,The renal protection of GLP-1 analogues on the DN is the dose-dependent.
