| Object The purpose of this study was to observation the change of Heart rate, Blood pressure, Cr, BUN, HSP70 of renal in the septic shock rat and to observation the interventional effects of ketamine.Methods The rat sepsis model was established by using cecal ligation and puncture (CLP). Fifty-four Male Wistar rats were randomly divided into SHAM group, CLP group and KT group. Before the operation of CLP, a blood pressure transducer were implanted into the common carotid artery for continuous registration of mean arterial blood pressure(MAP). After the operation, give sodium chloride 10ml·kg-1·h-1 till executed. SHAM group received laparotomy without any treatment, other groups underwent CLP group. To make a two-three centimeter cut in the middle of the anterior belly of rats, liberating the mesentery and cecum, using silk suture of 3-0 ligate the root of the rats' cecum, using the pin of 12 to make three holes in the root part of cecum. Two holes are separate from 3 centimeters, taking back the intestine, suturing the incision and closing the abdominal cut. And disinfecting the bandaging the cut. After the operation, giving sodium chloride 10ml/kg·h, till ending. In KT group at 20 min after CLP the intraperitoneal administration of 40mg/kg. Blood of heart was sampled for detection of renal function at 3h,6h and 12h after model induction. HE staining was used to observe pathological change of kidney.And the level of HSP70 was measured by immunohistochemical method.Results①General situation:Compared with the SHAM group, the activity of CLP group animal decreased, lethargy, piloerection, angle of eyes have Exudates and so on.The general situation of SHAM group is better.②The change of mean arterial blood pressure(MAP):after the model of CLP, the mean arterial pressure of all groups decrease gradually, the MAP of CLP group and KT group decrease significantly(P<0.01) after 9h; the MAP of CLP group achieve shock in standards; the levels of MAP in the KT group were significantly higher than that in CLP group(P<0.01), and no shock.③The level of BUN in CLP and KT groups raise strikingly at 6h and 12h(P<0.01), and the levels in the KT group were significantly lower than that in CLP group (P<0.01).④As compared with those of the SHAM group The level of Cr in CLP and KT groups raise strikingly at 12h(P<0.05), but the levels in the KT group were significantly lower than that in CLP group(P<0.05).⑤HSP70:HSP70 were observe in all groups. However, as compared with SHAM group, the expression of HSP70 increased strikingly in CLP and KT group (P<0.01). While KT group was obviously higher than CLP group (P<0.05).Conclusion During the septic shock the extent of HSP70 are higher Significantly, as the time prolonged. Ketamine can restrain sepsis rats blood BUN and Cr's heighten, relieve renal tissue pathology damage, for sepsis rats kidney has certain protection, the effect may be induced HSP expression related with enhanced. |
