Objective: Establish a rat model of Vascular dementia, in order to investigate the functional mechanism and repair effect on the Jiannao Yizhi tang of learning and memory, hemorheology, oxidative stress and pathological change of VD model rats.Methods: divide 60 rats randomly into 6 groups: sham-operated group, model group,donepezil, high,middle and low dose of Jiannao Yizhi tang group. Use 2-vo method to make VD rat model.. Each medicine group was lavaged with drugs or saline for 7 weeks. Use morris water maze test and step-down passive avoidance test to test learning and memory ability of the experiment rats. And then , we detected hemorrheologic indexes by taking brains , separated the precerebral cortex, to measure activity of MAO,CAT,GSH-PX , and compare the result with SPSS13.0.separating the left side hippocampus for hematoxylin and eosin staining.Through immunohistochemistry technique,we observe SS positive cells and GFAP positive cells.Results:1. The Morris water maze test and step-down passive avoidance task show the experimental rats'ability of learning and memory are different(P<0.05), while the ability increased significantly after treatment in each treated group comPared with model group(P<0.01 or P<0.05 ), moreover, there was no significant difference between 3 treated goups.2. Oxidative stress in the brain of model group rats was severe, meaning the GSH-PX activity was decreased in rats of model group(P<0.01), which was enhanced in rats of treated groups(P<0.01 or P<0.05).What's more, the activity of MAO,CAT,GSH-PX in donepezil group and approached to sham-operated group(P>0.05).3. SS positive cells were observed through immunohistochemistry technique shows that SS positive cells increase in drug group.Conclusion:1. The VD rat model established by using modified permanent bilateral common carotid arteries occlusion (2VO) basically simulated a clear cognitive impairment, increased blood viscosity ,oxidative stress, and ischemia injury of neurons etc. The rat model simulated well the state of chronic cerebral hypoperfusion of patients with VD .2. Jiannao Yizhi decoction improves the learning and memory ability caused by permanent bilateral common carotid arteries occlusion,and has similar effect to donepezil.3. Jiannao Yizhi decoction has strong function of anti free radical.4.Jiannao Yizhi decoction has the same function of alleviating ischemia injury significantly as Yangxue Qingnao granule,and the effectiveness is better when high does. 5.High and middle Jian nao yizhi tang show good result,but the low jian nao yi zhi tang has no different with model group.
