Objective:To observe the effects of clostridium butyricum on the expression of ITF, NF-κB and TNF-αin murine intestinal mucosa with ulcerative colitis and its possible mechanism.Methods:60 specific pathogen free, 6-8 week-old male BALB/c mice (weight 19.0±2.0g), wererandomly divided into 6 groups: normal, model, negative control, positive control, clostridiumbutyricum low-dose, clostridium butyricum high-dose group. 10 mice were involved in eachgroup. The normal group-mice were fed with distilled water. Intervention group-mice were fedwith 3% dextran sulfate sodium (DSS, MW: 36000-50000) freely for 7 continous days tocomplete ulcerative colitis modle. At the same time, except normal group, every exprementalgroup was orally administrated empty needle, physiological-saline, mesalazine (29.80mg/ml),clostridium butyricum (1×106CFU/ml), clostridium butyricum (1×107CFU/ml) respectively.During the exprement, general situation, daily weight, stool trait and occult blood were recorded,and mice were killed on the 8th day. Isolated the whole colon, and taken piece of colon formaldehyde fixed, paraffin embedded, Hematoxylin and Eosin (H&E) staining, the rest of thecolon were preserved in -70℃for production organization dissolving pulp.The following indiceswere used to assess the effect of ulcerative colitis model: disease activity index (DAI), colonlength, histological scores. Immunohistochemistry(IHC) was used to measure the expression ofITF and NF-κB in colonic mucosa. Radio-immunifaction method was used to measured theexpression of TNF-αin colonic mucosa.Results:1.BALB/c mice treated with DSS solution developed diarrhea, mucous stool and bloodystool.With days,DAI scores of all Intervention group were increasing. Clostridium butyricum canimprove the general situation of the mice.Compared with model group, the DAI scores, colonlength, histological scores were statistically significant(P<0.05).2.In positive control and clostridium butyricum groups,the expression of ITF was increased,the expressions of NF-κB and TNF-αwere both decreased, compared with model and negativecontrol group(P<0.05).The effect of clostridium butyricum low-dose group was samiliar tomesalazine . Conclusi:The manifestations of DSS-induced acute colitis mice were similar to clinical symptoms ofUC patients. Clostridium butyricum had therapeutic effects on the acute UC mice effectively. Itspossible mechanism may be related to the increase of ITF and the decrease of NF-κB and TNF-α.
