Objective:Pancreatic carcinoma is a kind of alimentary canal tumor with high mortality. Chemotherapeutics play a important role in its therapy. In this therapy,anti-cancer drugs which can kill tumor cells most but have the least toxicity to normal cells becomes one of the focuses. Allicin has many biological functions,such as anti-fungus,anti-bacterial function,decreasing blood fat and blood pressure, preventing from and treating arteriosclerosis et al.For the past years, antineoplasmic activities of Allicin have been paid closer and closer attention. We study inhibition function about Allicin to Pancreatic carcinoma cells and infection about Allicin to Pancreatic carcinoma cells cycle by MTT method and flow cytometric analysis.Methods:We used allicin in different concentration to interfere in PANC-1 cells which is in the exponential phase of growth in vitro.After Allicin treatment,we check cell Proliferation inhibition and IC50 of Allicin by MTT assay.The abscissa is Allicin consisteney, and vertical line is cell inhibitive rate, we can draw a chart about cell Proliferation inhibition, and chech IC50. Cells cycle were evaluated with flow cytometric analysis.Results:In this experimemt,we discovered:1. PANC-1 cells growth was inhibited by Allicin,and its IC50 of 24 hours was 5.45mg/L.2. These cells were treated by Allicin at the different concentrations of for 24 hours, compared with the control group, the Percentage of Go/G1 Phase of the cells was decreased and that of G2/M cells was inereased significantly in the group treated by Allicin (p<0.01),but the the Percentage of S Phase of the cells did not change.Conclusions:1.The proliferation of PANC-1 cells were significantly inhibited in a dose dependent manner after Allicin treatment.2.Allicin caused arrest of PANC-1 cells in G2/M phase.3.The study offered the experiment foundation in vitro for allicin to become the effective drug to treat Pancreatic carcinoma.
