Objective To investigate effects and mechanisms of Sulforaphane on the human glioma Cell (U251) apoptosis.Methods The inhibitory effects of sulforaphane on the growth of U251 cells were investigated by flow cytometric analysis and their value of IC50 were measured by 3-(4,5-dimethyl-thiazolyl-2)-2,5-diphenyl tetrazolium bromide (MTT) method. The expression of Cyt-c induced by sulforaphane was tested by Western blot analysis.Results Sulforaphane in low concentrations (≤100μmol/L) could significantly suppress the growth of U251 cells in a dose-dependent way. IC50 of U251 cell after 72 h incubation was (52.41±6.63)μmol/L. After 72 h incubation, The expression of Cyt-c showed obvious dose-dependency .Conclusions Sulforaphane could inhibit the growth of U251 cells and induce cell apoptosis. The induction of Cyt-c by sulforaphane mainly involved Mitochondrial way , which induces endogenous apoptosis.
