ObjectivesTo research retinoic acid and folic acid effects the proliferation, apoptosis on neural stem cell, and retinoic acid and folic acid differentiation the neural stem cells through the Notch signaling pathway for the central nervous system development, axon remodeling, nerve regeneration and nerve foundation for the treatment of diseases.MethodsWith different concentrations of retinoic acid stimulation of neural stem cells, in the process of adding folic acid intervention, to research the proliferation through immunofluorescence experiments on neural stem cell; use of DNA ladder, flow cytometry and TUNEL experimental techniques to observe the apoptosis on neural stem cell influence of retinoic acid and folic acid; using immunofluorescence, confocal, RT-PCR, real-time quantitative PCR technique to study the retinoic acid and folic acid on neural stem cells in Notch signaling pathway, and through this pathway abnormal differentiation of neural stem cells.Results1. Amount of retinoic acid had no obvious effect on the proliferation of neural stem cells, and adding folic acid can promote the NSC proliferation, and proliferation than the effect of folic acid alone was used.2. Excessive retinoic acid can cause apoptosis on NSC, but higher concentrations of folic acid can antagonize this effect the apoptosis rate of retinoic acid.3. 1μM retinoic acid can inhibit the expression of Notch signaling pathway molecules, causing NSC differentiate into neurons and glial cells, but can not differentiate into oligodendrocytes.4. Folic acid can inhibit or reduce differentiation through Notch pathway induced by retinoic acid.
