| Objective To study the effect of different doses of propofol preconditioning on myocardial energy metabolism in rats with Liver ischemia-reperfusion injuryMethods 48Wister rats were randomly divided into 4 groups each consisting of 12 rats :sham group(C group); ischemic reperfusion group(IR group );Low dose of propofol pretreatment group (BL group) High dose of propofol pretreatment group (BH group). Ischemic reperfusion models were made by ligated hepatic portal 30min .. The high energy phosphates(ATP,ADP and AMP) in myocardial tissue were measured by HPLC. MDA was measured by thiobarbiturate colorimetry,and SOD activity was detected by xanthine oxidase technique . The activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase of myocardial tissue were measured by chemical colorimetric methodResult the contents of ATP ,and EC in BL group and BH group were significantly higher than those in IR group,but lower than C group..The contents of ATP and EC in BL group were higher significantly than BH group.The content of MDA in IR group was higher significantly than that of C group,BL group and BHgroup.Besides, The activities of SOD in IR group was lower significantly than that of C group,BL group and BH group .The activities of Na+-K+- ATPase and Ca2+-Mg2+- ATPase in IR group were lower significantly than C group.The activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in BL group and BH group was higher than IR group,but lower than C group. The activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in BL group were higher significantly than BH group.Conclusion :propofol can protect ischemic reperfusion injury by raising the content of high energy phosphate in myocardium,meanwhile decreasing the content of MDA and recovering the SOD activities and the ATPase activities of myocardialt issue. And low dose group was better than high dose group... |
