Atherosclerosis is a kind of complex compound lesions. The main feature is its involved artery disease starts from endothelium, including have part of lipid and compound sugars gathered, bleeding and thrombosis, fibrous tissue hyperplasia and calcinosis, the gradual degradation and calcification of middle artery. Under normal conditions, endothelial cells can generate and secretion of various biological activity of substances, in vascular tension, growth, lipoprotein metabolism aspects plays a regulation function. But when atherosclerosis happens, vascular endothelial function is lower and disorder, appears endothelial dysfunction.Recombinant human Brain Natriuretic Peptide(rhBNP, xinhuosu) is to use genetic engineering technology of synthesis of BNP biological function identical with the restructuring of the product. At present it has been recognized and affirmed by expert in clinical practice of heart failure. Experiments have shown the artery of BNP can prevent cattle vascular endothelial injured, BNP unique mechanism may make treatment atherosclerosis into a new area, so reversal endothelial disfunction is the new trend of cardiovascular disease treatment.Purpose:The effects of Brain Natriuretic Peptide on the function of rats arteriosclerosis model vascular endothelial. Further defined mechanism of action of rhBNP against AS. Provide scientific theory for the clinical treatment of atherosclerosis disease.Method:40 healthy wistar rats, divided into five groups at random, eight rats in contorl group, eight in model group, each of the treatment group of nitrates(Hua ren xin shu), Sodium Ferulate (Haisi),rhBNP (xinhuosu) model group were eight rats.Model and the treatment group were given a high-fat feed, the control group give ordinary feed, free of water. After 12 weeks broken tail picks, centrifugal separation out serum, ultra-low temperature (-80℃) freeze-stored.(?) The treatment group were given nitrates and ferulic acid sodium, restructuring the brain natriuretic peptide subcutaneous injection, the control group and the model group gives saline subcutaneous injection, ten days later take heart blood from sudden death rat, centrifugal separation serum, all specimens to collect after reunification detection. Adopt enzyme-linked immunosorbent adsorption (ELISA) methods to determine each rat before and after the treatment serum TXB2 and 6-Keto-PGFla content, meanwhile determination of conventional assay inspection ((?) Blood triglyceride and cholesterol levels, etc), and light-microscopy before and after the treatment of rats aortic endothelium cells changes. Before and after the treatment groups compares the sample mean differences of matching t test,(?) each outcome using multiple independent sample mean differences F text, statistical analysis.Consequence:a. There was no statistically significant difference compare with each rat general data (P>0.05). b. High-fat feed each routine laboratory inspection comparison there was no statistically significant difference (P> 0.05). c. The treatment group after medical treatment, serum TXB2 and 6-Keto-PGFla levels than before treatment difference obvious, statistically significant (P< 0.05). d. Before and after the treatment of each group drug therapy in serum TXB2 and 6-Keto-PGFla content is difference. but has no significant difference(P>0.05).e. Optical observations hints, the control group vascular lumen has big plaques,no calcium sediment.Elastin membrance pigmented can easily be seen.Tunica elastica is made of smooth muscle cells,arranging disorganized.The smooth muscle cell nucleolus is boom and ectoblast is conjunction tissue.The treatment group is smoother than model group, local endometriosis has a few MC adhesion, visible EC missing, but better than model group, and between treatment groups has not seen the obvious difference.Conclusion:rhBNP can not only through increased the content of PGI2 arterial stiffness rat models, lower TXA2 level, but also in morphology can reduce atherosclerosis rats damage, vascular endothelial protecting endothelial function, but also improve the arterial blood supply. For clinical further treatment atherosclerosis disease provide theoretical basis.
