Backgrounds and Objeetions: Hypertensive disorders in pregnancy is a disorder specific to pregnancy and a major cause of maternal and perinatal morbidity and mortality.However,The etiology and pathogenesis of hypertensive disorders in pregnancy have not been clarified.Many evidences suggest that hypertensive disorders in pregnancy would be associated with many facts,such as lipid metabolic disorder ,Vascular endothelial cell injury,inflammatory reaction and insulin resistance .APN has effects that including increasing insulin sensitivity,protecting endothelial cells,anti-inflammatory and anti-atherosclerosis.So APN becomes one of important factors to be studied on hypertensive disorders in pregnancy.Materials and Methods:Placental and basal decidua tissues were collected from normal pregnancies(n=18),mild preeclampsia(n=10) and severe preeclampsia(n=12).The expressions of APN and its receptors(AdipoRl/2) were detected by using immunohistochemitry in the two tissues respectively. Integral optical density(IOD)was detected by image analysis system(Image-proplus 5.0),which was represented the strength of the expressions of APN and its receptors.The IOD was took to be statistically analyzed by analysis of variance(ANOVA)and t-test used SPSS17.0 system. The aim is to analyse the association between the expressions of thses three indexes mentioned and hypertensive disorders in pregnancy.Results:The expression of APN in placenta was low in the placenta of hypertensive disorders in pregnancy. Statistically analysed,there was significant difference of the expression of APN in placenta between in normal pregnancy and in mild preeclampsia and severe preeclampsia,respectively(p>0.05),and there was more markedly lower in severe preeclampsia than that in mild preeclampsia(p>0.05).The same results were showed in basal decidua tissues about the expression of APN in hypertensive disorders in pregnancy.The changes of the expression of APN in placenta and basal decidua in hypertensive disorders in pregnancy were coincidental, and the two expressions were positively related.Both of the receptors were presented in placenta and in basal decidua tissue,with adiponectin receptor 2 expressed mainly.There was no significant difference of the expression of adiponectin receptor 1 among groups in both of the two tissues. However, the expression of adiponectin receptor 2 rised with the aggravation of hypertensive disorders in pregnancy.The expression of adiponectin receptor 2 in placenta in severe preeclampsia was significantly higher than that of in normal pregnancy and in mild preeclampsia,respectively(p>0.05),also the same result in basal decidua tissue.Conclusions and innovation:APN expressed in both placenta and basal decidua ,which stated that the APN in maternal-fetal interface was of great significance to maintain physical pregnancy. The expression of APN in maternal-fetal interface was low in hypertensive disorders in pregnancy,and gradually reduced with the aggravation of hypertensive disorders in pregnancy.The APN in maternal-fetal interface may be involved in the pathogenesis of hypertensive disorders in pregnancy, further research about the expression of APN in the interface will help reveal the pathogenesis. The expression of APN between in placenta and basal decidua has positive correlation.Both AdipoR presented in maternal-fetal interface, with AdipoR2 expressed primarily.AdipoR2 was rised markedly with the aggravation of hypertensive disorders in pregnancy.Maybe,the APN in maternal-fetal interface play its roles through AdipoR2.Hypertensive disorders in pregnancy is influenced by both the maternal and fetal sides effects. It is unilateral to neglect either of the factors. In the past , the study ignoring maternal uterine decidua basalis, focused much concentration on placenta.This experiment for the first time makes the trophoblast in maternal-fetal interface and decidua basalis together as a whole, on which we study the adiponectin and its receptors ,exploring interaction with hypertensive disorders in pregnancy. It expands a new field in the gestational hypertension disease's research.
