Objective:To investigate the expression of chemokine SDF-1 in inflammatory cells from bone marrow and its receptor CXCR4 in tumor cells and their prognostic value in esophageal cancer.Methods:The expressions of SDF-1 in inflammatory cells from bone marrow and CXCR4 in tumor cells were detected by S-P immunohistochemistry in 116 Specimens of the esophageal cancer tissues. The relationship of their positive expression to the prognosis of esophageal cancer patients was analyzed.Results:The three-year survival rate of all patients is 63.79%(74/116).SDF-1 is expressed mainly in cytoplasm and cell membrane in inflammatory cells from bone marrow in esophageal cancer tissues. There are no significant difference in three-year survival rates between SDF-1-positive and SDF-1-negative patients. The positive expression of SDF-1 are found to have no influence on survival of esophageal cancer patients (p>0.05).CXCR4 is expressed mainly in cytoplasm and cell membrane in tumor cells in esophageal cancer tissues. The three-year survival of CXCR4-positive patients is significantly lower than CXCR4-negative patients.The positive expression of CXCR4 are found to have significant influence on survival of esophageal cancer (p <0.05), it is negatively correlated with survival period. CXCR4 positive expression is higher in patients with lymph node metastasis than those without(p<0.05), and is also higher in T3-4 stage than T1-2 stage in the invasion depth (p<0.05). Both SDF-1 and CXCR4 expressions are stronger inâ…¢-â…£stage than inâ… -â…¡stage (p< 0.05). Multivariate analysis indicate that pTNM stage and CXCR4 expression is the independent prognostic factors in esophageal cancer patients (p<0.05).The expression of SDF-1 in inflammatory cells from bone marrow show no significant relationship with the expression of CXCR4 in tumor cells. Conclusion:SDF-1 is expressed mainly in cytoplasm and cell membrane in inflammatory cells from bone marrow in esophageal cancer tissues.these inflammatory cells included plasma cells, lymphocytes and neutrophils. SDF-1 is expressed strongly in most plasma cells, and moderately in lymphocytes, whereas only weakly expressed in sparsely appeared neutrophils. CXCR4 is expressed mainly in cytoplasm and cell membrane in tumor cells in esophageal cancer tissues.Positive expression of CXCR4 in esophageal cancer is the independent prognositic factor. The increased number of positive cells is related to poor prognosis.The expression of SDF-1 showed no significant relationship with prognosis.
