The nerve regeneration and functional recovery after Spinal cord injury (spinal cord injury, SCI) has been a medical problem, nearly years, in cell transplantation, with its unique biological characteristics and the role of promoting the axons after spinal cord injury, olfactory ensheathing cells (olfactory ensheathing cells, OECs) become the most promising treatment methods. Currently, the main manner in treatment of spinal cord injury with Olfactory ensheathing cell transplantation is local transplantation, and there is no way to retrieve relevant research reports about intravenous transplantation in the treatment of spinal cord injury.ObjectiveExplore and study the effect(sensory motor function recovery, the regeneration of nerve fibers, etc.) and the possible mechanism about olfactory ensheathing cells transplantation for acute spinal cord injury, and provide a simple method of transplant for the treatment of acute spinal cord injuryMaterials and methods1. culture, purification and identification of Olfactory ensheathing cell:Take olfactory bulb of healthy wistar rat two months or so, irrespective of gender, the body weight is about 200-250g,culture, purify the primary OECs, then digest and collect the olfactory ensheathing cells, and mark nuclear with diphenyl azomethine.2. Preparation and transplantation in the treatment group of Spinal cord hemisection injury model:40 healthy adult Wistar rats, prepared by the right side of T10 spinal cord hemisection injury model. The rats were randomly divided into the olfactory ensheathing cells transplantation group of local intramedullary (A group), olfactory ensheathing cells transplantation group (B group), D/F12 vein graft group (C group) and control group (D group). 3. the evaluation of Spinal cord injury functional recovery:score limb motor function respectively (improvement CBS ratings) by 1 day, 1 week,2 week,3 weeks,5 weeks and 7 weeks; 10 weeks after transplantation animal model of motor function observed situation; After five weeks,10 weeks of spinal cord derived samples, first, morphology in general, then HE staining and silver staining, to observe the regeneration of parts of axons in spinal cord injury, and the survival, distribution of labeled cells in the spinal cord injury with immunohistochemistry and fluorescence microscope.ResultsApplication of differential adhesion and Ara-c inhibition of purified culture method combined with high purity cultured olfactory ensheathing cells, cell body is three forms, spindle-shaped, flat round and multiple ecphyma,and NGFRP75 and GFAP staining is positive reaction.The period 1-10 weeks after transplantation,the motor function of rat hindlimb can recover in varying degrees,the 10 week, A, B, C, D Group CBS score was 8.2±1.874,14.60±3.273,35.90±2.234,35.80±4.290.3 weeks after the score shows, B group and A group with the C group and D group compared with the time difference was significant (P<0.01), B group and A group of similar efficacy, C and D group were no significant differences (P> 0.05).We see there were more regenerated axons in A group, B group by HE staining and silver staining,,and the nerve fibers through the injury area, the axon regeneration of A group is more than B group, C group and D group only a few scattered regeneration of axons, no continuity of nerve fibers through the injury area. Immunohistochemistry can be observed in the damage zone survival of olfactory ensheathing cells, distributed in the damage zone, and in the survival of the quantity of group A more than B group.Conclusion1. Differential attachment and Ara-c inhibition of purified culture method combining the advantages of both integrated,and can be to a higher purity of olfactory ensheathing cells, it is simple, effective and practical application;2. Treat spinal cord injury with Olfactory ensheathing cells transplantation, its efficacy and interlocking partial transplant is similar to a certain degree of impaired neurological function recovery, and transplantation of olfactory ensheathing cells can survive in the spinal cord area and migration; 3. Olfactory ensheathing cells transplantation operation is simple and can be repeatedly treated ,and can avoid re-operation caused by a number of spinal cord secondary injury and postoperative complications,it has been better clinical practice and value.
