ObjectiveObserved the esophageal squamous cell carcinoma (ESCC) whether to exist in vasculogenic mimicry(VM) this special blood supplying mode, and through the examination hypoxia inducible factory-1α(HIF-1α) expression in the ESCC, analysis of the meaning between HIF-1αand VM, to investigate HIF-1αand the occurrence of VM.MethodsCollect 170 ESCC cases with complete clinical pathology data, immuno-histochemical staining of CD34 and PAS special histochemical staining were conducted to observe if VM exists in ESCC;and by SP immunohistochem-ical detection of HIF-la protein expression, and counted MVD,SPSS13.0 statistical software was used for data processing.Results(1)Conducted on CD34 and PAS double staining,and combined with HE staining,vasculogenic mimicry exists in esophageal squamous cell carcinoma. The typical forms of VM are vessel-like structure which formed by tumor cell without staining CD34,and the PAS-positive looping pattern,tumor cells are separated from the tubes by PAS-positive matter like basement membrane,in which red blood cells presented.Near the structures,there were no hemorrhage, necrosis and inflammatory cells infiltrating.(2)VM was observed in 21 cases of 170 ESCC samples (12.4%), VM positive rate of ESCC was significantly higher than in normal esophageal mucosa (P<0.01), and with differentiation,lymph node metastasis, TNM stage, MVD related (P<0.05), but it had nothing to do with ESCC patient's age, the sex (P>0.05). VM in poorly differentiated esophageal squamous cell carcinoma was higher than that in moderately to well differentiated;VM with lymph node metastasis was higher than that without lymph node metastasis;The stageⅢ-Ⅳof VM was higher than that in the stageⅠ-Ⅱ;The VM-positive group was lower MVD than the VM-negative group.(3) There are 109 cases of ESCC tissues with the existence HIF-1αprotein expression (positive rate 64.1%)in 170 cases,100 cases of normal esophageal mucosa in 13 cases only the existence of HIF-1αprotein expression (positive rate 13.0%).ESCC of HIF-1αprotein expression was significantly higher than the corresponding normal esophageal mucosa, a statistically significant difference between the two groups (P<0.01).It was proved having no statistica-lly relationship with age,gender,differentiation (P>0.05),but having statistically positive relationship with lymph node metastasis, TNM stage, MVD.HIF-1αwith lymph node metastasis was higher than that without lymph node metastasis in 170 cases of ESCC; The stageⅢ-Ⅳof HIF-1αwas higher than that in the stageⅠ-Ⅱ;The HIF-1α-positive group was higer MVD than the HIF-1α-negative group.(4) VM and positive expression rate of HIF-la protein in ESCC were showed statistically positive correlation(P<0.05).Conclusion1.Vasculogenic mimicry existed in esophageal squamous cell carcinoma, which was the special blood supplying mode. It correlated with differentiation, lymph node metastasis,TNM stage and MVD.2.HIF-1αprotein was highly expressed in ESCC,which correlated with lymph node metastasis,TNM stage and MVD.3.Overexpression of HIF-1αinduced the formation of VM channels.
