Adenine phosphoribosyltransferase is a one of the key members of purine which catelizes adenine to AMP. According to the reported.SjAPRT1 and SjAPRT2 gene sequences (GenBank Accesion:AAW24796,AAW25340) we cloned and expressed the two isoforms of SjAPRT. Realtime PCR and western blotting were used to investigate the different transcription level and expression of SjAPRTs gene during the developmental stages.The two genes were subcloned into pET-28a plasmid and the recombined plasmids were transformed into E. coli BL21, The recombinant protein were expressed and purified by Ni-NTA agarose column. And the protective efficiency of rSjAPRT1 was tested in immunized mice. In humans, the rSjAPRTl antibody level was evaluated by ELISA in sera from 20 non-infected humans and 50 schistosomiasis patients.RT-PCR results showed specific bands of SjAPRT1 and SjAPRT2 transcripts existing in cercariae, eggs, schistosomulum and adult worms. Realtime PCR revealed the transcription level vary during the different developmental stages, the transcription level of adult worms and eggs were twice of that in schistosomula. Western blot showed that SjAPRT1 existed in egg, schistosomula and adult worms but not cercariae, while SjAPRT2 was detected in cercariae, schistosomula and adult worms but not egg and the specific bands in adult is weak. So the two isozymes of SjAPRT may play roles in different developmental stages of schistosoma and SjAPRT1 maybe play important role in adult worms. While SjAPRT2 may play role during the junior stages. At the same time, we evaluate their poteintial application on diagnosis and vaccine.The recombinant proteins could be specifically recognized by the S. japonicum infected rabbit sera. The ELISA results of rSjAPRTl and rSjAPRT2 showed a highly significant difference between the two groups with a sensitivity of 60%,63.3% and a specificity of 96.6%,93.3%, The worm reduction rate and the egg reduction rate of rSjAPRT1 were 13.6% and 14.4% respectively.
