Objective:To investigate the activation of autophagy lysosomal pathway in rat hippocampal neurons induced by ischemia-hypoxia/reperfusion and study the role of autophagy activation in neuron death.Methods:In this study, we established primary cultured rat hippocampal neurons as ischemia-hypoxia model in vitro. After 2 hours oxygen-glucose deprivation and different times of reperfusion (OGD/R) treatment in primary cultured hippocampal neurons, neuron viability was evaluated by MTT assay, the specific structure of autophagosome was visualized by transmission electron microscopy, and the autophagy-specific protein microtubule-associated protein 1 light chain 3 B (LC3B) was detected by immunofluorescence. The inhibitor of autophagy 3-Methyladenine (3-MA) was also used to exam the viability of neurons.Results:Treatment by OGD/R markedly reduced neuronal viability (P |
