| Objective:To investigate the effect of high volume hemofiltration (HVHF) treatment on expression of triggering receptor expressed on myeloid-1 of peripheral blood monocytes and its functions in patients with sepsis.Methods:Twenty patients with sepsis were involved in this prospective, matched pair study.10 patients were treated with high volume hemofiltration, and another 10 patients were as matched pair without high volume hemofiltration treatment。(1) TREM-1 mRNA level were determined by RT-PCR at 0 st and 72th post-treatment hour in HVHF group and matched pair group. (2) Peripheral blood monocytes were isolated in vitro at 72th post-treatment hour, and incubated in the culture plate。The monocytes were stimulated by LPS at 6th post-incubation hour。The expression of TNF-a and IL-6 in the supernate were measured by ELISA at 48th post-incubation hour.Results:(1) The expression of TREM-1 mRNA were significantly lower in HVHF group compared with matched pair Group,0.604±0.078 vs 0.871±0.061, p<0.05. (2)The expreesion of TNF-αand IL-6 in the supernate were markedly lowerer in HVHF group with matched pair Group, p<0.05.Conclusions:HVHF treatment can markedly down-regulate the expression of TREM-1 in peripheral blood monocytes, let it maintain at an appropriate level, and maybe reduce an excessive inflammatory response. |
