The natural active substances of resveratrol from the peanut root was extracted and separated by the ultrasonic extraction, macroporous resin purification and Atmospheric silica gel column chromatography purification, and especially the process conditions were studied systemicly.(1) Through the quadratic general rotary unitized design, ultrasonic extraction mathematical simulation model was concluded by the lin-ear regression analysis:y=0.078X,+0.033X2-0.030X3-0.075X4+0.055X1X2+0.060X2X3+0.056X2X4+ 0.036X3X4-0.133X12-0.074X22-0.075X32-0.073X42+3.922, Then the optimum conditions of ultrasonic extraction peanu-t resveratrol was determinded by the mathematical statistical analysi-s:ethanol concentration is 73%, ultrasonic power is 290 w, extraction time is 5.6min, ratio of material to liquid is 17.5:1, Its extraction ra-te on theory which was calculated by mathematical simulation model can be achieved 3.96%o. The theoretical model was proved that was reliable by the verification experiments.(2) After the type optimized test of macroporous adoption resin, DA-201 resin adsorption kinetics experiments, DA-201 resin adsorption isotherm testing, the loading volume test and the dynamic eluting test and the column chromatography experiments on three factors(loading velocity, eluting velocity and concentration of eluting solvent)that can be concluded:DA-201 resin adsorption process for the resveratrol conformed to Freundilch and Langmuir equation, and the process of absorption was heat absorption, but is physical adsorption process; the optimum conditions of DA-201 macroporous resin column chromatography test was determined by these experiments and all kinds of analysis:the concentration of sample is 0.7mg/mL, pH= 3, the sample volume is 20 mL, elution volume is 15mL, macroporous resin purification resveratrol mathematical model can be achieved:y=75.516-4.790X1-1.549X2+8.914X3-1.612X1X2-2.542X12-2.498X22-8.107X32Through Regression analysis and Variance analysis, the optimum conditions on macroporous resin purification were drawn:the sample velocity is 1.0 mL/min; the elution flow is 1.6 mL/min, the ethanol concentration is 76%, the yield rate of purified resveratrol is 80.43±0.13%, which was measured by HPLC, and the purity of separated resveratrol can reach 39%.(3) Through the TLC, silica gel column chromatography and HPLC purity experiments, chloroform and methanol in the ratio of 9:1 as the eluting solvent, and the silica gel adsorbents and sample ratio of 50:1 were optimum conditions for the Atmospheric silica gel column chromatography. With the silica gel column chromatography the purity of purified resveratrol powder by the macroporous adoption resin can be advanced from 39% to 99%.
