Nicotine is present in the Solanaceae (Solanum) of the alkaloid, is also an important component of tobacco. When nicotine enters the human body, will have many roles: as nicotine limb peripheral vasoconstriction, heart rate, blood pressure, breathing fast, mental status change (such as emotionally stable or mental excitement), and to promote platelet aggregation, is caused by heart blood vessel blockage, hypertension, stroke and other cardiovascular diseases, the main accomplice. Nicotine addictive or produce dependence, re-use nicotine also increased heart rate and elevate blood pressure and reduce appetite. In addition, studies have shown that nicotine can directly affect sperm production, hinder spermatogenesis and maturation.Embryonic stem cells (ESCs) are from the mammalian blastocyst inner cell mass during the (InnerCellMass) of the separation, and able to maintain the undifferentiated state in vitro, and can be highly proliferation of pluripotent cells, which has unlimited in vitro proliferation, self-renewal and differentiation characteristics. Either in vitro or in vivo environment, ES cells can be induced to differentiate into almost all the cells in the body type that developed into embryos after the biological systems can be simulated body cells and tissues of complex interactions between, which in the field of drug research a wide range of uses. The best use of embryonic stem cells are differentiated into various types of cells, for "cell therapy", for cell transplantation offers the immunogenicity of the material, any disease involving the loss of normal cells to pass through the transplantation of embryonic stem cell differentiation and by the to the specific cells to treat. Such as the use of nerve cells to treat neurodegenerative diseases(Parkinsons disease,Huntingtons chorea,Alzheimers disease,etc.),with islet cells to treat diabetes,with cardiac and other cells to repair myocardial necrosis.In this paper, we used the classical method of myocardial cells, the mouse embryonic stem cells into embryoid (embryoid bodies, EB s), then access to myocardial cells by inducing differentiation. The establishment of normal culture negative control, normal myocardium induced positive control and different concentrations of nicotine in the experimental group, after 2 weeks of induction, RT-PCR results showed that compared with the positive control group, nicotine promotes embryonic stem cell dry gene Oct-4, Rex-1 expression, inhibition of the cardiac differentiation marker genes a-actin, desmin, cTnI expression, suggesting that nicotine promote proliferation of mouse embryonic stem cells while inhibition of myocardial differentiation of certain.
