Objective To explore the possible physiological impacts of cell line NIT-1 attributing to high concentration Free Fatty Acids by laser tweezers raman spectroscopy.Methods Cell line NIT-1 was cultured in RPMI 1640 supplemented with 0.25mmol/L Palmacid for 0, 12, 24, 48 hours respectively, one group of cells served as control and the cells were incubated without any free fatty acid. Raman spectra were obtained by the mean of scanning combined of Hoechst33342/PI fluorescent staining. The spectral difference was analyzed by OriginPro8.0.Results Raman spectra analysis shows that the shift changes of Raman peaks at 780cm-1, 1124cm-1, 1092cm-1 and 1172cm-1 and the peaks intensity decline at 780cm-1,1172cm-1,855cm-1 and 873cm-1, while there was no obvious difference between control groups(p<0.05). These peaks showed above mainly correspond to intracelluar DNA and protein.Conclusion The high concentration Free Fatty Acids highly relate with NIT-1 cell physiologic function impaired and apoptosis. Raman spectra have the advantage of observing cellular structure changes in early period.
