Objective: To study sperm morphology changes before and after the density gradient centrifugation and swim-up method processing, analysis if there is a correlation between the sperm morphology and sperm DNA damage, and whether the processed sperm morphology, sperm DNA damage affect assisted reproductive technologies (ART) end and discuss the correlation.Method: Data from 2010 September to November of 51 infertility couples who took the first IVF treatment in our center, routine examination of man semen were studied respectively before and after density gradient centrifugation and swim-up method .Sperm morphology using Diff-Quick analysis of strict sperm morphology. Processed semen samples using sperm chromatin diffusion (SCD) method for detection of sperm DNA integrity. Observe the fertilization on the first day after in vitro fertilization and cleavage on the third day. Results: After the density gradient centrifugation and swim-up method processing the normal form sperm significantly increased, middle deformity sperm rate and mixed deformity sperm rate has declined obviously than treatment before (P < 0.05). Head deformity sperm rate and sperm DFI positively correlated, difference was statistically significant (P < 0.05), and normal sperm forms, middle sperm deformity rate of deformity sperm rate, the rear rate, mixed with sperm deformity sperm rate though DFI negatively correlated, but the difference was not statistically significant (P > 0.05).There were no significant correlation between the normal form sperm rate, head deformity sperm rate, middle deformity sperm rate, tail deformity sperm rate and ART outcome, P>0.05.Sperm DNA fracture index (DFI)and fertilization rate ,zygote transgne rate,2PN transgene rate are tending to a significant negative correlation (P<0.05), and both optimal embryo rate,2PN transgene rate also negative correlation , but this was not statistically significant (P>0.05).Conclusions: The density gradient centrifugation and swim-up method treatments can reduce middle deformity sperm rate and mixed deformity sperm rate, significantly improve the normal form sperm rate; Head deformity sperm rate and sperm DFI positively correlated; The damage of sperm DNA has an effect on the treatment of IVF. When the DFI of sperms increases, the fertilization rate of IVF decreases. And then has an effect on the development of fertilized eggs'cleavage and embryos. Sperm DNA integrity can be a detection index of Sperm quality,and predict the effect of IVF.
