1.The expression of P—JAK2,P—STAT3 and TGF—p1 of renal tissue in the diabeticnephropathy rats in different period.Objective:To establish model of diabetic nephropahic rats and to observed p-JAK2,p-STAT3 and TGF-B1 expression in diabetic rat glomeruli in different periodMethords:32 male wistar rats were randomly divided into normal control group(N group) (n=16),diabeticnephropathic model group(DN group)(n=16).The two grop had two sub-groups according to different time:4w,12w,8 rats in each group We established the rat model of diabetic nephropathy induced by streptozotocin(STZ)and right nephrectomy All the rats received daily gavage for 12 weeks Observed the blood sugar,urinary protein/24h Rats in control group and model group for 4 weeks were sacrificed at the end of 4 weeks Rats in control group and model group for 12 weeks were sacrificed at the end of 12 vc-eeks respectively We also observed the renal pathological changes by optics microscope mad transmission electron microscope Expression levels of p-JAK2,p-STAT3 and TGF-B1 in renal tissue of rats were detected by immunohistochemistryResults:Pathological changes such as thickening of glomeruler basement membrane,foot process with disorder and proliferation of mesangium matrix that characterise the diabetic nephropathy has been found in DN model group at the end of 4 vc-eeks by optics microscope and transmission electron microscope These changes such as basement membrane thicken,foot process fusion,increased mesangial matrix were much more severe in renal tissues of DN group rats at the end of 12 weeks p-JAK2,p-STAT3 mad TGF-B1 expression level ofthe renal tissue of rats of DN model group is higher than the N group at the same period(respectiVely,P<0.05).The expression ofp-JAK2,p-STAT3 and TGF-B1 ofrenal tissue in the diabetic nephropathy rats of DNgroupatthe end ofl2weekswere highterdistinctlythanthose ofDN groupatthe end of4 vc-eeks (pO.05).and pathology changes were not be found in the two control groupsConclusion:The result suggests JAK/STAT signal transduction can be concern with the progress ofthe diabetic nephropathy2.Effect of Yishen Capsule on activation of JAK/STAT signal transduction of Renal Tissue in the Diabetic Nephropathy Rats.Objective:To investigate the effect of Yishen capsule on activation of JAK/STAT signaltransduction of renal tissue,the blood sugar,Urinary protein/24h and the renal pathologica chmages in the diabetic nephropathy rats and to explore possible mechanisms on kidney protection of Yishen capsuleMethods:Wistar rats were randomly divided into 4 groups:the normal control group(N group),the diabetic nephropathy group(DN group),Yishen capsule treated group mad the losartan treated group,10 rats in each group The rats models of diabetic nephropathy were established successfully.The rats in Yishen capsule treated group were given Yishen capsule 625 mg/kg through intragastric administration once daily for 12 weeks,the rats in losartan treated group were given losaactan 30 mg/kg through intragastric administration once daily for 12 weeks During 12 weeks,Levels of blood sugar,quantities of 24 hours urinary protein excretion were detected All rats were sacrifced at the end of 12th week,patho-logical changes on renal tissue have been examined and observed,the expression of the p-JAK2,p-STAT3 and expression of TGF-B1 was measured by immunohistochemistry in the kidneyResults:In 12w,compared with those in the normal control group immunohistochemistry noted that the expression of p-JAK2,p-STAT3 protein and TGF-B1 protein in the kidney were significantly increased in diabetic nephropathy rats and two durg treated group(pO.05)Conclusion:The result suggests Yishen capsule can ameliorates renal injury via the inhibition of activation ofJAK/STAT signal transduction and the relevant TGF-B1 expression in the kidney of diabetic rat...
