PartⅠEffect on the different concentrations of oxidized LDL on THP-1 cell apoptosisObjective To investigate the effect on the different concentrations of ox-LDL inducing THP-1 cell apoptosis. Methods THP-1 cell were induced by PMA for 48 hours to become macrophages.Then macrophages incubated and treated with different concentrations(0 to 100μg/ml) of ox-LDL for 48 hours,and with 75μg/ml ox-LDL for different time (0~72h). Experiment was divided into 3 groups : control group,nLDL group,ox-LDL group (different concentration,different time ). The degree of THP-1 cell apoptosis was determined by flow cytometry. Results Incubation THP-1 cell with ox-LDL, the apoptosis of THP-1 cell were increased with a dose-dependent and time-dependent effect. The apoptosis rate increases as the concentration and dose increases. Conclusions: Ox-LDL can induce THP-1 cell apoptosis with a dose-dependent and time-dependent effect .PartⅡOx-LDL through P38MAPK signaling pathway induce THP-1 cell apoptosis and related mechanism analysis on the endoplasmic reticulum stressObjective To investigate the endoplasmic reticulum stress mechanism of ox-LDL inducing THP-1 cell apoptosis and do further research on its effect mechanism.. Methods THP-1 cell were incubated and treated with 100μg/ml ox-LDL for 48 hours in experiment and divided into 3 groups at random: control group, ox-LDL group, ox-LDL + SB group.Then Western blot and RT-PCR were used to examine the expression of GRP78,Caspase-12 ,CHOP and P38MAPK . Results Compared with untreated cells, The level of GRP78 ,CHOP, P38MAPK protein and mRNA had increased significantly in the ox-LDL group and ox-LDL+SB group(p<0.05). However, ox-LDL+SB group could decrease the level of GRP78 ,CHOP, P38MAPK protein and mRNA compared with ox-LDL group(p<0.05).Conclusions Ox-LDL can induce endoplasmic reticulum stress in educational THP-1 cell and SB can release endoplasmic reticulum stress injury on the apoptosis of cell through AMPK pathway.
