Cervical cancer is one of the most common malignancy of women, second only to breastcancer in second place,threat to quality of life of women.Tumors is the result of multiplefactors of angiogenesis, invasion and metastasis, cell proliferation, malignant transformation andso on. Our previous study had selected the differential expression gene by cDNA gene chip toEarly stage cervical squamous and found the expression of MIF with lymph node metastasedwas higher, so we speculate MIF Plays an important role in Cervical cancer invasion andmetastasis.Purpose: By observing whether MIF can regulate expression of vascular-related factors IL-8and VEGF through the ERK / MAPK signal transduction pathway,we can discover preliminarythe mechanism of affecting the cervical cancer angiogenesis and metastasis in early stagecervical cancer.Method:firstly, MIF,ERK1,IL-8,VEGF of vascular-related factors protein expression wasexamined with Immmunohistoehemieal analysis,Western blot analysis and ELISA .Secondly, Recombinant human MIF on the proliferation of cervical cancer cell lines wasexamined by MTT, MIF,ERK1,IL-8,VEGF of vascular-related factors protein expression afterstimulated by Recombinant human MIF was examined with Immmunohistoehemieal analysis,Western blot analysis and ELISA .thirdly, ERK inhibitor PD98059 on the inhibition of cancer cell lines was examined by MTT,MIF,ERK1,IL-8,VEGF of vascular-related factors protein expression after inhibited by wasexamined with Immmunohistoehemieal analysis,Western blot analysis and ELISA .Finally, MIF,ERK1,IL-8,VEGF of vascular-related factors protein expression afterstimulated by the interaction of ERK inhibitor PD98059 and Recombinant human MIF wasexamined with Immmunohistoehemieal analysis,Western blot analysis and ELISA.Result: MIF,ERK1,IL-8,VEGF of vascular-related factors is highly expressed in cervicalcancer, The expression of CaSki that is Transfer of intestinal cells is higher than Siha, It isshowed that the expression of these factors may be closely related to the metastatic potential ofcells. Recombinant human MIF stimulates the expression ,ERK inhibitor PD98059 inhibites theexpression, ERK1,IL-8,VEGF of vascular-related factors protein expression was inhibited bythe interaction of ERK inhibitor PD98059 and Recombinant human MIF, We can speculate thatMIF may be explored through the ERK/MAPKsignal transduction pathway for expression ofIL-8 and VEGF, and promot angiogenesis. ConclusiConclusion: MIF related to vascular-related factors such as IL-8,VEGF can promoteangiogenesis, cell proliferation and thus promote the growth and metastasis of cervical cancer.Through stimulation of Recombinant human MIF and inhibition of PD98059, we may guess MIFcan regulate expression of IL-8 and VEGF through the ERK / MAPK signal transductionpathway. But PD98059 did not completely block the expression of these factors, we speculatedthat there are other pathways.By blocking this pathway, We can inhibit cell growth andangiogenesis of cervical cancer. The metastasis mechanism of MIF may be explored through theERK/MAPKsignal transduction pathway in early stage cervical cancer.
