bjective:To investigate the effection and mechanism of fasudil,the inhibitor of Rho kinase,on erectile function of hypertensive rats.Methods 15 healthy male 12-week-old Sprage-Dawley rats were randomly devided into 3 groups(five rats for a group):control group(group A),hypertension group (group B), fasudil treatment group (group C).Establishment of the renal hypertention model with Renal artery ligation.4 weeks after surgery, measuring the systolic blood pressure of rats tail,which are higher than 160mmHg are successful models.And the rats are measured systolic blood pressure for every 3 weeks. Group C is received intraperitoneal injection of fasudil(30mg/kg/day),group A and B are given intraperitoneal injection of normal saline. After surgery of 10 weeks, measure the rat corpus cavernosum pressure/mean carotid arterial pressure (ICPmax/MAP),measure the ROCK1 protein, ROCK2 protein expression levels in the corpus cavernosum with Western-blot。Results:The systolic blood pressure,the expression of ROCK1 and ROCK2 protein in group B were significantly increased (p<0.05) than group A.The systolic blood pressure,the expression of ROCK1 and ROCK2 in group C were significantly reduced (p<0.05) than group B. ICPmax/MAP of group B significantly decreased than that of group A(p<0.05),group C significantly increased than group B(p<0.05). Conclusion: Fasudil may improve erectile function in rats with renal hypertension by means of inhibiting the expression of the RhoA/Rho kinase signaling and decreasing the blood pressure.
