| Objective:Detect the expression of LIFR in different phases of infantile hemangiomas,vascular malformation and normal skin to investigate the mechanism of proliferation and regression in infantile hemangiomas. Methods:The tissue of vascular lesion was obtain from 83 patient who were underwent surgery from 2009-2010. We get 30 samples from proliferating phase of infantile hemangiomas (Group A),22 samples from involuting phase (Group B),16 in vascular malformation and 15 in normal skin samples. All the samples were classified by the criterions of Mulliken. The expression of LIFR,CD133 and Ki67 were examined in normal skin,vascular malformation and different phases of infantile hemangiomas with SP method of immunohistochemieal technique. Then divide the results into 3 levels:negative (-), weak positive(+) and strong positive (++) according to the two factors: expression intensity and the ratio of positive cells.The data we have collected were dealt with soft SPSS17.0, and less than 0.05 are considered significant. Results:1. The positive expression of LIFR is that brown particles were mainly on the cytomembrane and cytolymph; In proliferating phase, the positive rate were 76.67%(23/30); In involuting phase, the positive rate decline to 31.82%(7/22); In vascular malformations and normal skin the positive rates were 25%(4/16) and 20%(3/15); It was significantly higher in proliferating phase than in involuting phase,vascular malformation or normal skin.2. The positive expression of CD 133 is that brown particles were mainly on the cytomembrane and cytolymph; In proliferating phase, the positive rate were 60%(18/30); In involuting phase and vascular malformation, the positive expression were 9.09%(2/22) and 6.25%(1/16); Between the proliferative phase and the involuting phase, the difference was statistically significant (X2=13.898, P<0.05).3. The positive expression of Ki67 is that brown particles were mainly in nucleus; The positive ratio of proliferating hemangiomas was 86.67%(26/30), involuting hemangiomas was 27.27%(6/22), the difference was statistically significant (X2=9.816, P<0.05); And in other two groups only very weak expression were detected.4. The expression of LIFR has correlation relationships with CD 133 and Ki-67, strong positive expression mainly assembled in proliferating phase under the 6 months, weak positive were present in proliferating phase of over 6 months old children or in early involuting phase. In 52 cases of hemangioma, there are 8 cases that LIFR and CD 133 were both strongly positive,6 cases were both weak positive,20 cases were both negative; and there are 11 cases that LIFR and Ki67 were both strongly positive, 9 cases were both weak positive,17 cases were both negative; By spearman's rank correlation, there are positive correlation relationships among the expressions of LIFR,CD 133 and Ki-67 in infantile hemangiomas, and the correlation coefficients were 0.63 and 0.787 (P<0.01). Conclusions:1. Overexpression of LIFR in proliferating hemangioma, suggest that LIFR may correlate with the development of infantile hemangiomas.2. CD133 positive endothelial progenitor cells have participated in the occurrence and development of hemangioma. 3. The expression of LIFR has a positive correlation with CD 133 and KI67 in hemangioma. It may conclude that these three factor have some influence on each other, and they also have some synergistic action in development of hemangioma. 4. The difference that LIFR,CD133 and KI67 expressed in different phase has a great benefit to discuss the treatment and prognosis of hemangioma. |
