Objective:To explore the source of dendritic cells (DC) in central nervous system after interacerebral hemorrhage (ICH) and its injury mechanism,by produced the intracerebral hemorrhage model of rats caudate nucleus and examination reation of the change of the brain edema,the expressions of interleukin-6(IL-6) and granulocyte macrophage-colony stimulating factor(GM-CSF) with the OX62+ OX42+cells surrounding hematoma at different time. Methods:1. Autologous blood was injected into the caudate nucleus of rat that produced intracerebral hemorrhage model.2.50 SD rats were randomly divided into 2 groups which were sham operation group and ICH group. There were 25 rats in each group, the animals in each group were divided again into 24h,48h,72h,5d and 7d after the model was produced. There were five animals at each time point.3.The models were evaluated at 24h post the operation. Neurological dysfunction score was assessed at 24h,48h,72h,5d and 7d post the operation.4.Observed water contents of brain tissue arounding the hematoma at different time points by wet-dry-weight method.5.Measured GM-CSF and IL-6 contents in the brain tissue arounding the brain hematoma at different time points by enzyme-linked immunosorbent assay (ELISA).6.The change of DC and neuroglial cells arounding the hematoma in each group were observed at the corresponding time points by immunohistochemistry and microscope techiques. Results:1.The sham operation group didn't take place obvious neurological dysfunction at each time points. The ICH group took place the most serious neurological dysfunction at 48h, then was lower. The ICH group took place more serious neurological dysfunction(P<0.05)at each time point than the sham operation group.2.The water content of brain tissue arounding brain hematoma of ICH group had not statistics obviously (P>0.05) compared with the sham at 24h, Then the ICH group began to ascend higher and 48h-72h its peak, had statistic significance obviously (P<0.05) compared with the sham. Then the ICH group has descend at 72h after the surgery, and though still more 5d-7d the higher compared with the sham but there were no statistics differences (p>0.05).3.The IL-6 contents arounding brain hematoma has to ascend higher compared with the sham at 24h, had statistic obviously (P<0.05), and 48h its peak, had statistic obviously (P<0.05),then has descend, the IL-6 contents in the brain tissue homogenates of the ICH and sham had no statistics differences (p>0.05)at 7d after the surgery. The GM-CSF contents arounding brain tissue had no statistics differences (p>0.05)at 24h compared with two group. Then the ICH group began to ascend higher at 48h, had statistic obviously (P<0.05) compared with the sham.and still higher, had statistic obviously (P<0.05) still 7d.4.The OX62+OX42+cells and neuroglial cells had obvious changed occur in the ICH at each time points. The neuroglial cells had began to ascend higher compared with the sham at 24h, had statistic obviously (P<0.05), and still higher,7d at its peak, had statistic obviously (P<0.05).; The OX62+OX42+cells had began to ascend higher compared with the sham at 24h, had statistic obviously (P<0.05), and still higher,7d at its peak, had statistic obviously (P<0.05).5.Linear correlation:The OX62+OX42+ cells of ICH group was positively correlated with the GM-CSF contents arrounding brain hematoma of ICH (R>0,P<0.05); The OX62+OX42+ cells of ICH group was positively correlated with the neuroglial cells arounding brain hematoma of ICH (R>0, p<0.05); The IL-6 contents in the brain tissue homogenates of ICH group was positively correlated with the water contents of brain tissue arounding the hematoma(R> 0,P<0.05).Conclusion:1. The ICH model of rat was made by autologous blood that is simple,make easily and the cheaps. The models are as almost same as the ICH of patients, thus can be used generally.2. Observed the DC of surrounding brain hematoma after ICH,which probably was one of the mainly derived from the neuroglial cells surrounding hematoma.3.The role of DC surrounding brain hematoma depended on its environmental conditions. The DC may aggravating brain damage.In early of hematoma,by promote inflammation; and promote the damage tissue repaired in the later,by suppression immune response,...
