Objective:To investigate the protective effects and mechanism of Catalpol and puerarin freeze-dried powder for injection (CPFPI) on PC 12 cell injure induced by CoCl2.Methods:Different concentrations of CoCl2 were used to induce the cytotoxicity in PC 12 cells in order to optimize concentration by MTT assay. PC 12 cells were pretreated with or without different concentrations of CPFPI for 1 h and then exposed to CoCl2. MTT assay was used to explore cell survival; the levels of SOD and NO were assessed with colormetric microassay. In addition, Caspase-3, Bax, Bcl-2, IκBαexpressions were detected with Western blotting.Results:After treatment with 500μmol·L-1 CoCl2 in PC 12 cells for 24 h,the cell viability markedly decreased about 60%. Compared with blank group, each dose group on PC 12 cells survival activity has no remarkably difference (P< 0.05). Compared with model group, CPFPI significantly increased the cell viability in a dose-dependent manner in PC12 cells injured by CoCl2 (P< 0.05, P< 0.01). When further investigate the mechanisms of CPFPI on CoCl2 induced apoptosis of PC 12 cells, we found that CPFPI (24.50,49.00,98.00μg·mL-1) significantly increased SOD levels, and (12.25, 24.50,49.00μg·mL-1) significantly decreased NO content. In addition, CPFPI can significantly decrease the expression of Caspase-3 protein, improved the expression of IκBαprotein, as well as elevated the ratio of Bax to Bcl-2, by regulating Bcl-2 family protein expression (P< 0.01. All these suggested that CPFPI was a potential neuroprotective agent and its neuroprotective effects were achieved at least partly by promoting endogenous antioxidant enzymatic activities, inhibiting free radical generation, activation of caspase-3. and regulation of Bcl-2 and IκBαConclusions:CPFPI strongly protects PC 12 cell injure induced by CoCl2,and inhibits the apoptosis of the injured PC12 cells。...
